Ion chromatography has been one of most important and effective tools for detection the ionic compounds with the 40 years’ development. Therefore, it widely applies to energy, chemical industry, biological medicine and food, which provide accurate data for directing production and supporting the scientific research. However, with the development of expanding in the field of ion chromatography, the diversity and complexity of the samples have become more and more. So, how to test accurately and analyze of unknown material under complex matrix situations is an important issue in the development of ion chromatography.In this study, ion chromatography were applied to test the unknown stances microbial cultures with the method of valve switch online pretreatment. And the valve switching and other separation conditions were optimized to determine and quantify the analyzed material. The main findings are as follow:1. The developments, principle, classifications and main application of ion chromatography have been introduced. We also specially described the progress of the pretreatment and their applications.2. 17 kinds of organic acid and 7 kinds of anion were simultaneously determined by using conventional single column ion chromatography and electrical conductivity detector. Meanwhile, we accurately tested the target substances with this method. This method has been applied to simultaneous determine organic acids and inorganic anions in microbial cultures with satisfactory results.3. The automatic switching capillary ion chromatography system coupled with two valves and two pumps were established. Therefore, many analytical conditions were optimized. Trace pyruvate was accurately detected in cyanobacteria culture using this new method.
