Affinity membrane chromatography is a novel technology combining membrane technology and chromatography. Nanofibers have been paid more attention as affinity membrane because of their advantages, including high specific surface area and outstanding transport property. Solution blowing is a new nanofiber fabrication method which have recently received increasing attentions. Meanwhile, solution-blown nanofibers are commonly curled in three-dimension and this configuration enhances their transport property.Chitosan is widely used in affinity adsorption, however, it’s hard to spin with pure chitosan and it’s mechanical properties is poor. In this study, coaxial solution blowing process was applied to spin core-shell nanofibers, with chitosan/PVA as shell, nylon 6 as core and EDGE as the crosslinking agent. This structure combines the utilization of adsorption materials of the chitosan layer with the mechanical properties improvement because the PA6 layer material.The morphology of the nanofiber mats was observed using field emission scanning electron microscopy (SEM) and the core-shell structures of the fibers were confirmed through transmission electron microscopy (TEM). F3GA (CB) was then immobilized onto the membrane surfaces for subsequent protein affinity adsorption and confirmed the presence of CB molecules on the nanofibrous membranes by FI-IR. At last, BSA adsorption capacities of the CB attached to the chitosan/PVA-nylon 6 nanofibers were measured, and the BSA adsorption capacity achieved by the NCNF membranes at the ratio of core/shell is 1:1, chitosan:PVA=50:50 and pH 7.14 with immobilized CB was 379.43 mg/g with fast adsorption rate, and the adsorption isotherms accorded with Langmuir equation.This paper provides a certain theoretical basis for the core-shell composite nanofibers applying to separation and purification of protein, which offers a new way for affinity membrane separation.
