This article mainly focuses on Peony leaf, the total acid total phenol total sugars and total protein in Peony leaf are determined, several different methods are employed for the extraction, separation and purification of total flavones, then the anti-oxidation capacity of the different extracts are determined respectively.First, the content of total acid and phenol total sugars, protein in Peony leaf are found to be, 1.623%, 3.653%, 24.569% and 16%. So it is clear that the macro molecular substance takes up a great proportion in Peony leaves,some proper method can be chosen to remove the impurities laying a foundation for separation and purification of flavonoids from leaves of Peony. respectively, laying a foundation for separation and purification of flavonoids from leaves of Peony.Second, by the methods of single factor and orthogonal experiment, I find the optimum process for the extraction of Peony leaf flavonoids by microwave and ultrasonic to be:1. miscrowave time 20min, ethanol concentration 70%, the ratio of solid to liquid 1:12.2. the optimum process for ultrasonic extraction of total flavone: temperature 60°c, ultrasonic time 20min, ultrasonic power 600W, the ratio of solid to liquid 1:25.The two methods are compared in terms of extraction rate and yield, the method of microwave-assisted extraction have an edge over the ultrasonic extraction, eventually the total flavonoid content reaches 1.97%, and purity reaches 10.56%.Then, the macroporous adsorption resin and ultrafiltration combination methods are combined in different orders to purify the flavonoids of microwave-assisted extraction. In this article, I use D-101 macroporous adsorption resins and molecular weight for 10000Da interception of polyethersulfone ultrafiltration membrane, compare the results of the two purification methods to determine the best process for purification of total flavone of Peony. Finally, the lard and sesame oil are analyzed qualitatively and quantitatively,the higher of the purity of flavonoids from peony leaves explain the more obvious effect in the peony. the auther draw the conclusion that the higher purity ,the better anti-oxidation capacity ,which means that flavonoids from leaves of Peony play a important role in the the anti-oxidation capacity.
