In this paper, like mung bean waste as raw material was prepared with the enzyme function of lowering blood pressure ACE inhibitory peptides, the use of membrane technology for the initial ACE inhibitory peptide separation and purification; macroporous resin ultrafiltration through static adsorption and desorption of liquid , the dynamic adsorption and desorption, and further purified ACE inhibitory peptides1,Selecting neutral protease, alkaline protease, pepsin protease, papain protease to make enzyme experiment for mung bean protein, to rate the degree of hydrolysis and ACE inhibition index, the results show that the hydrolysis degree and ACE inhibition of alkaline protease was higher than that of other protease . After the microwave heat treatment mung bean protein of enzyme hydrolysis than the effect of enzyme hydrolysis, pretreatment of mung bean protein in the optimal solution by microwave power is 500W, the time for the 30s.With single factor experiments and response surface method , we can know the optimal hydrolysis conditions for alkaline protease: substrate concentration 2%, pH7.5, hydrolysis time 120min, 3% enzyme, hydrolysis temperature of 55℃; alkaline protease optimal conditions , the ACE inhibition is 84.83%, the molecular weight distribution is wide.2,With ACE inhibition rate of flux and the indexes to compare four different molecular weight cutoff ultrafiltration membrane on membrane flux and the impact of ACE inhibition, the results show that the 3000 MWCO membrane flux is relatively large, ACE inhibition have a higher rate. By single factor experiments and orthogonal test the optimal membrane parameters: feed temperature 35, ultrafiltration time, 10min, ultrafiltration pressure 0.08MPa, pH8, in this condition, the average flux reached 32.71%. After ultrafiltration, molecular weight of 3000 to collect the following effluent, ultrafiltration permeate most of the molecular weight has been below 700, the income component of the ACE inhibition is 87.32%.3,Compared nine macroporous resin adsorption properties of mung bean peptides, experiments show that the adsorption resin DA201-C better performance on the adsorption rate of mung bean peptide 70.45%. By DA201-C macroporous resin can effectively take off in the ultrafiltration permeate salt concentration is 30%, 50%, 70%, 90% ethanol eluted solution through ultrafiltration and found that the higher through the liquid by ultrafiltration DA201-C treatment with macroporous resin with high concentrations of ethanol elution fractions after the relatively high blood pressure activity in the eluted fractions in the graded ethanol by 70% eluted fractions of ethanol the highest ACE inhibitory activity, ACE inhibition rate of 91.23%, compared with UF permeate increased 5.78%.
