In this paper, firstly the research progress of the reaction of the primary amine groups of amino acid and protein with epoxides were summarized. Secondly, the primary amine groups content of collagen peptide were determined. Thirdly, effects of time, pH, temperature and feed ratio on the reaction of allyl glycidyl ether with collagen peptide were investigated and orthogonal test was designed based on single factor analysis. Lastly, the kinetics of reaction of collagen peptide with allyl glycidyl ether was studied by developing a kinetic model. The main contents of this paper are as follows:(1) The determination of the primary amine groups content of collagen peptideâ‘ The veracity of Van Slyke's apparatus was validated by determining the primary amine groups contents of glycin and glutamic acid.â‘¡The percentage of collagen peptide was calculated from the nitrogen content of gelatin determined by the Kjeldahl method.â‘¢The primary amine groups content of collagen peptide was determined by the Van Slyke method.(2) The conversion was determined by the Van Slyke method, colorimetry and gravimetric analysis, respectively. Effects of time, pH, temperature and feed ratio on the reaction of allyl glycidyl ether with collagen peptide were investigated and orthogonal test was designed based on single factor analysis.â‘ Four factors were investigated with the conversion of the primary amine groups as index.â‘¡The conversion of the primary amine groups was determined by the Van Slyke method. The conversion of AGE was determined by colorimetry and gravimetric analysis, respectively. The quantity of AGE which reacted with other active groups of collagen peptide was calculated by combining the Van Slyke method with colorimetry. The error in determining the conversion of the primary amine groups by gravimetric analysis was calculated by comparing the three methods.â‘¢The test was designed according to L25(45) of orthogonal test. The reaction time, the pH level, temperature and material ratio were optimized and the optimal reaction conditions was obtained.(3) The kinetics of reaction of collagen peptide with allyl glycidyl ether was studied by developing a kinetic model. â‘ Taking AGE 30 times the dosage of the primary amine groups, the variation law of the conversion of the primary amine groups was studied by monitoring the reduction of the primary amine groups as a function of time. The reaction order with respect to the primary amine groups was determined by applying the kinetic model.â‘¡Taking AGE 10, 15, 20, 22, 25 times the dosage of the primary amine groups, the relationship between AGE concentration and the conversion of the primary amine groups was established and the reaction order with respect to epoxy groups was obtained.â‘¢The reaction rate constants were determined at different temperatures(30, 35, 40, 45 oC)and the activation energy was calculated.
