N-(2-hydroxy)propyl-3-trimethylammonium chitosan chloride (HTCC) with well water-solubility was prepared by modifying the chitosan(CS) with glycidyl trimethyl ammonium chloride (GTMAC). HTCC and poly (vinyl alcohol) (PVA) and its blend films were prepared by aqua-solution method. The blend film was crosslinked using glutaraldehyde (GA). The thermo properties of films were tested by the thermogravimetic apparatus (TG). And Fourier transform infrared spectroscopy (FTIR) was applied to characterize the structure of neat PVA, HTCC and their blend films. The gel content, antibacterial activity and swelling behavior in buffer solutions of pH value 7.4 at 37℃were investigated for PVA-HTCC blend films in details, meanwhile, the effect of HTCC content on the property of films and the mechanism of film crosslinked by GA were also discussed. Results of TG showed that the blend films have better thermo property than the neat PVA and HTCC films. The equilibrium swelling degree(ESD) of the blend films increased with the content of HTCC, it was up to 212% when the HTCC content reached up to 40 (wt)%. The gel content of films depended on the content of HTCC and GA, which increased with the decreasing of GA content, and this might be the main reason of the ESD of blend films increasing. The antibacterial activities against Staphylococcus aureus and Escherichia were studied for the blend films. Results showed that the antibacterial rate of un-crosslinked blend film exceeded 99%, even though the antibacterial activity of blend film was weakened slightly by crosslinking, it still showed a superior antibacterial efficacy. Finally, the mechanism of PVA-HTCC blend film crosslinked by GA was discussed according to the analysis of testing results. Besides, the blend mixture of PVA/HTCC was used for electrospinning to investigate the spinnability, nanofiber nonwoven membranes were obtained and showed in the scanning electronic microscope (SEM) images, which can be seen that the diameter of nanofiber and membrane pore-size were evenly distributed. This proved the good spinnability of PVA/HTCC blend. And these results indicated that the high potential applied possibility of HTCC/PVA blend in biomedical field.
