In this paper, the common components and composition of amino acids of several kinds of umbrella and oral arms of fresh and pickled jellyfish were detected and analyzed firstly. They had approximate composition in the Rhopilema esculenta and Stomopholus nomurai. The protein of Rhopilema asamushi and Rhopilema hispidium were less than Rhopilema esculenta. The total amino acids in the umbrella of Rhopilema esculenta was 127.32 mg/g by dry weight. The content of essential amino acids was as high as 29.31%.Two determination methods of ACEIP were compared at present. The UV spectrophotometer method had more disadvantages, such as badly repeatability and low accuracy. For so many uncontrollable factors and too much manipulation, the method wasn't used. With some simple operation it needn't extract hippuric acid in the reactive solution by visible spectrophotometer method, and it showed good repeatability and high sensitive. However, the data were unreliable by using crude extracts from lung of pig, only to use standard could get precise results.The condition of mensuration of thin layer chromatography (TLC)-spectrophotometer method was studied to determine ACEI's activity. The precision coefficient of variation was 2.17% (n = 8). With this method it had better repeatability and stability data when determining ACEIP. However, by this method operation was complicated and steps was cumbersome, also more influence of human factors. The thin layer chromatography method for establishing was simple and high accurcy when determining, the linear regression equation: y = 20431.098 +713.059 x, the correlation coefficient was 0.99185. Determination precision of coefficient of variation was 0.04 percent.To make ACEIP from jellyfish with the method of enzymolysis, the results of process parameters was that added neutral protease by 50ku/100gPr, and enzymatic hydrolysated 9h in water of 45℃after washing, homogenate and sterilization. The samples which went through 5kDa ultrafiltration membrane after rough filtration could go with maximum ACE inhibition rate. The samples which were concentrated at temperature of 100℃had little influence on inhibiting ACE's activity, but freezing could make its inhibitory rate significantly reduced.The anti-hypertensive effect of ACEIP on renal arteries stenosis caused hypertension model and L-NNA induced hypertension model in New Zealand white rabbits was studied. The results showed that the jellyfish ACEIP exerted the anti-hypertensive effect 0.5h after the experimental animals were administered with the sample by gavag. When the dosage was less than 60mg/kg.bw, the anti-hypertensive was positively correlative with the dosage. An increased dosage resulted in a longer action time, but could not lower the blood pressure below the normal value. The blood pressure of normal rabbits was not influence by ACEIP from jellyfish.The anti-hypertension effects of five different ACEIP samples from jellyfish in vivo and in vitro were compared, and the results were similar. The anti-hypertensive effect of the samples after freezing treatment were significantly reduced, but sterilization and concentration had little effect on the anti-hypertensive effect. Simulation of digestive action in vitro had little influence on jellyfish ACEIP. The result indicated that ACEIP from jellyfish was anti-digestive. Samples treated by debittered and sustained-release were contrast in vitro and vivo, and the result indicated that the best processing method was that: added 0.5% powdered activated carbon first and keeping 20min at 55℃, then added 1.5%β-cyclodextrin at 65℃and embedding 30min. It was not only removed bitter, but also greatly prolonged the efficacy and play a role of sustained-release effect.ACEIP from jellyfish was contrast with four chemically synthesized peptide drugs, In vitro tests it showed: Activity Inhibition rate of Beijing No.0 was lowest; Benazepril's inhibitory rate reached 64.9%, and it was maximum; ACEIP from jellyfish was followed by Benazepril; Valsartan and nifedipine had no inhibitory effect. The effect in vivo test: with the same dosage, anti-hypertensive effect of nifedipine on hypertensive model was not obvious by nifedipine which was calcium antagonists; The action time of jellyfish ACEIP was faster than valsartan to achieve the same effect, but it maintaining short time. Added dosage the jellyfish ACEIP could come at the same effect with valsartan. Compared with hydrochloride-benazepril, benazepril had larger amplitude of anti-hypertensive,but adding dosage would cause hypoteinsion. So that big fall of blood pressure changing was unfavorable for patients with hypertension. However, this thing would not happen with jellyfish ACEIP.The expansion of process of jellyfish ACEIP was studied, and anti-hypertensive effect had been determined in vivo and in vitro. Sample D9 and Sample E10 both could make hypertensive value fell to normal value in vivo, and it could keep 20 hours. The ACE activity inhibitory rate of D9 and E10 were 67.3% and 61.5% respectively in vitro. But the other samples were lower.
