Androstenedione is a key intermediate of microbial steroid metabolism. It can be produced by microbial side chain cleavage of phytosterol, however this method still has the difficult of substance transferring,it makes the transforming rate still low. In this article, we focus on the effects of protamine , glycine, surfactants and ultrasound in improving the fractional conversion of androstenedione at the basis of preliminary studies. The research also optimize the fermentation conditions, it provides a theoretical basis of microbiological method for industrial production of androstenedione. The main conclusions of this paper are as following:By the study of inhibitors of cell wall structure, we confirm the use of glycine, when the concentration is 3.0g/L, the product of the concersion rate is 83.7%. By the study of surfactant, we confirm the use of T-80/S-80, when the concentration is 0.100%, the product of the concersion rate is 85.6%. By the study of ultrasound, we confirm using the ultrasound irradiation 60W ultrasonic 180s every 12h can make the product of the concersion rate increase to 85.3%. By ultrasonic irradiation with T-80/S-80 synergy study, we can see that when T-80/S-80 concentration of 0.075% every 12h ultrasound 150s, the product of the conversion rate up to 88.7%At the basis of the above-mentioned study, through the orthogonal test to optimize the fermentation tank to be the optimum conditions are as follows: the initial pH value of 8.5, temperature 32℃, Ventilation per liter fermentation broth IL/min, stirring rate 450r/min, Under these conditions the product of conversion rate up to about 86.6%. On this basis, the study of fermentation broth changes in pH values ,it found that at 60-80h fermentation broth the pH values reduce from 6.9 down to 5.7, test proves that even if the fermentation time to 60h every 12h regulation of pH value of a conversion of a product beneficial to the conversion rate up to 89.7%, the fermentation cycle shortened to about 96h, in-depth study we found that add an appropriate amount of sterile water to the fermentation broth in fermentation process is good at the transformation of product.This article also examined the two-phase system Androstenedione the extraction process,with the adoption of the technology of extraction of the product, the extraction rate could reach 76.61%, the product purity reach 98.24%.
