Simultaneous distillation extraction(SDE) is a novel sample preparation method for extracting,isolating and concentrating volatile and semi-volatile compounds in complex sample. It is often used as the pretreatment method prior to GC or GC-MS, and widely applied in the analysis of volatile components in food,tobacco and flavors. In the present study, we optimized the conditions of SDE. Under the optimum operating conditions, we gained the satisfactory reproducibility. In addition, we applied the developed method to the analysis of tobacco from different location, and it may establish the foundation for tobacco fingerprint.In the first chapter, different pretreatment methods for complex sample are reviewed, including water stream extraction(WSE), simultaneous distillation extraction (SDE), ultrasound-assisted solvent extraction(UASE), solid-phase microextraction(SPME) and supercritical fluid extraction(SFE). The basic principle and main working parameters of simultaneous distillation-extraction (SDE) were introduced in detail, and its applications in the field of food, plant essential oil, tobacco, and so on are summarized.In the second chapter, ultrasound-assisted solvent extraction(UASE) and simultaneous distillation extraction (SDE) were compared for their effectiveness in the extraction of volatile compounds from tobacco. The results showed that SDE gained the higher effectiveness. The main working parameters (solvent, distillation-extraction time, weight of sample and salting-out.) of simultaneous distillation-extraction (SDE) were studied. Under the optimum operating conditions, satisfactory reproducibility was achieved.In the third chapter, the volatiles of tobaccos from different source (Yunnan, Henan, Anhui, Sichun, Chongqing, Shanxi, Guizhou, Heilongjiang, Huhan, Hubei, and so on) are analyzed by SDE-GC/GC-MS. In total, 150 componets are identified, such as Linalool, Phenylethyl alcohol, Megastigmatrienone, Tetradecanoic acid, n-Hexadecanoic acid, 9,12-Octadecadienoic acid (Z,Z)-, and so on.
