Therapy of maxillofacial region malignancy is an existent tough problem of oncotherapy. It is for a long time that the therapy of the head and neck tumors mainly depends on the surgery operations. The blood supply of orai and maxilliofcial part is flourish and the anatomic stucture is complicated,so the conventional surgery has some problems such as high risk,heavy trauma,high relapse rate ,et al. Besides the features deformation after operations and malfunction has badly affection on patients's life value. The method of transcatheter arterial embolization(TCAE ) combine chemotherapy offers a new approach for therapy of the oral and maxillofacial mailgnant tumors.Choice of embolic chemotherapeutic agent is a critical role on intervention chemoemoblization therapy. It has been a hot spot on embolic praeparatum. Gelatine is a base material that commonly used in prepare microballoons. Glutaraldehyde usually used as cross-linking agent which could not be cleared thoroughly in gelatin-microballoons and has toxic effect on tissues, that would confine clinical application of gelatin-microballoons. The study is to choose polyaldehyde dextran as a new kind of crosslinking agent to prepare gelatin-pingyangmyc(PYM)-microspheres ,it could significantly reduce toxic effect on tissues because used glutaraldehyde as cross-linking agent and be applied securely intervention chemoemoblization therapy.Objective:To choose a new kind of crosslinking agent to prepare gelatin—Pingyangmyc(PYM)-microspheres for the interventional chemoemoblization therapy.Method:.This study is divided into two parts: first,we prepared gelatin-PYM- microspheres cross-link with polyaldehyde dextran and glutaraldehyde respectively by emulsified cold一condensation method; second,we compared morphology and biological characteristics of two kinds of microsphere by related empirical method.Result : The roundness of polyaldehyde dextran group is better than glutaraldehyde group though optical microscope; the distribution range of particle diameter of two kind of microsphere were fit for the use of embolization, however, polyaldehyde dextran group is narrower when compared with glutaraldehyde group(p<0.05);the drug content rate and the encapsulation rate of polyaldehyde dextran group was 94.6 % ,8.9 % respectively which was higher than glutaraldehyde group 80.1%,5.5%(p<0.05);the cumulative release rate of PYM from oxidized-dextran group in 6 hours was 71% , it has lower drug release than glutaraldehyde group 95%(p<0.05). MTT experimental results show that polyaldehyde dextran group has higher cell survival rate than glutaraldehyde group.Conclusion: These results implied the polyaldehyde dextran as a new kind of crosslinking agent has more exerted quality than glutaraldehyde,gelatin-PYM- microspheres cross-link with polyaldehyde dextran could become a perspective granular pattern embolism for targeted tumor therapy.
