The extraction of ecdysterone(ECD) and total phytosterone from Adiyranthes bidentata with different methods was studied in this paper, and optimized the extraction technics conditions with different optimizing ways.In the process of supercritical fluid extraction ECD from Achyranthes bidentata, the flow rate of CO2 was 2L/min, static extraction time was 10min. The kind of modifier, flow rate of modifier, extraction pressure and temperature were studied first, the preliminary optimum conditions were: the flow rate of modifier (methanol) 0.5mL/min, extraction pressure 250bar and the extraction temperature 70℃. With designing the orthogonal array design on L9(3)4 matrix, the optimum technics conditions were: the extraction pressure 250bar, extraction temperature 70℃, flow rate of modifier(DMSO) 0.5mL/min, dynamic extraction time 1h. Each of the four parameters had the similar effects on the results, so there was no significant parameter.In the process of ultrasonic extraction ECD and total physterone, the steady conditions were: the herb was sieved through a 250μm filter, extraction frequency 40kHz. The preliminary conditions: the extraction time 20min, 60% ethanol, the ratio of liquid and material 10:1. With response surface methodology, the results of ECD were: extraction time 26min, 65% ethanol, the ratio of liquid and material 12:1; total physterone: extraction time 26min, 55% ethanol, the ratio of liquid and material 8:1.In the experiment of separation ECD and total physterone with macroporous resin, the optimum conditions achieved with "step-by-step" method were: the concentration of ecdysterone in loading sample 94.976μg/mL, total physterone 2.6417mg/mL, the volume of loading sample 5BV, loading flow rate 0.6BV/min, the eluant was 70% ethanol, eluating volume 5BV, eluating flow rate 0.8BV/min.
