Genistein(5,7,4'-trihydroxyisoflavonid), a flavonoid isolated from soy beans. It is potential antibacterial, antioxidant, anti-inflammatory and anticancer agent. Herein, genistein-Cr(Ⅲ) complex was synthesized and characterized, and its free radical scavenging activity, the binding mode and affinity to calf thymus DNA were investigated. This thesis is composed of four chapters:In chapter 1, the structure of DNA, the mechanisms of metal complexes with DNA and the used methods of the interaction between metal complexes with DNA were described.In chapter 2, the Cr(Ⅲ) complex of genistein (Gen-Cr) was synthesized and characterized by UV, IR, MS, elementary and thermogravimetric analyses. The results suggested that the complex has the constituent: Cr(C15H9O5)3·2H2O.In chapter 3, the free radical(·OH, O2-, DPPH·) scavenging activities of the Gen-Cr complex were investigated. The results showed that free radical scavenging activities of the Gen-Cr complex on·OH, 02- and DPPH. were obviously stronger than that ofgenistein.In chapter 4, the mechanism of the interaction between the Gen-Cr complex with calf thymus DNA (ctDNA) was investigated by using means of UV spectra, fluorescent spectra and viscosity. The results indicated that the Gen-Cr complex should bind ctDNA mainly by intercalation. The binding constant of complex with ctDNA was determined. The experimental results suggested that the Gen-Cr complex maybe a promising candidate for anticancer, which deserves further research.
