The aim of study is to optimize the extraction process of Chidan pill and to prepare a better effective and convenient drug.In addition,an effective Chinese traditional medicine according with the good extraction process was provided for women chronic pelvic inflammatory disease.MethodsThe orthogonal design was used to select the optimum extract of CDP according to the contents of Peoniflorin and TanshinoneⅡA.The three factors and three levels of orthogonal design were determined whether they had some influence on the extraction process.Alcohol quantity and water quantity,extracting time,alcohol concentration were evaluated which would be the basis of selecting the optimum extraction conditions.RP-HPLC was used to measure their contents of Peoniflorin and TanshinoneⅡA.The method of measuring the content of Peoniflorin is that Agilent Eclipse XDB-C18 column (4.6×150mm,5μm)was used,the mobile phase was acetonitrile-water-phoshporic acid(17:83:0.1) , detection wavelength was 230nm, the column temperature was 30℃.The method of measure the content of TanshinoneⅡA was that Agilent Eclipse XDB-C18 column(4.6×150mm,5μm)was used,the mobile phase was methanol-water (75:25) , detection wavelength was 270nm and the column temperature was 25℃.The models of mice ear tumefaction caused by xylene and rat foot tumefaction caused by carrageenan were used to observe the anti-inflammatory action of OECDP via oral administration. The effect of OECDP on relieving pain was tested by counting body twisting induced by glacial acetic acid and measuring the latent period of licking hind foot with the hot plate method in mice.By using the Phenol Glue, the Wistar big mice were made to have chronic pelvic inflammatory disease as experiment.Observe the pathologic picture of the mice;detect the radio immunoassay to examine respectively the content of serum (IL-2) and Tumor necrosis factor.Observe the effect of treatment of OECDP.ResultThe optimal preparation process was as follows: the mixture of medical materials was firstly refluxed with total 10 times of 70% alcohol, 1.5 hours and then extracted with total 10 times of water, 1.5 hours.Alcohol concentration affected the contents of Peoniflorin and TanshinoneⅡA significantly while the boiling time had no obvious effect.The test of pharmacodynamics indicate:the drug could restrain mouse ear tumefaction and rat foot tumefaction, decrease mouse body twisting number in glacial acetic acid model and increase the pain threshold with hot plate method.OECDP can significantly improve indexes of hemorheology.Extent of pathological changes significantly decreased.The serum TNF-αlevel in the model group were significantly higher than those in the normal group and the OECDP group;while the IL-2 levels in model group were significantly lower than those of other 6 groups.Conclusion1.The optimal preparation process was as follow:the mixture of medical materials was firstly refiuxed with total 10 times of 70% alcohol, 1.5 hours and then extracted with total 10 times of water, 1.5 hours.The optimal preparation process is reasonable with high rat of active component which can provide reference for preparation of CDP.2.In the CPID model rats induced by phenol glue,OECDP can improve the correlative parameters,which indicted that OECDP possess protective effect in vivo; immunoregulation for CPID model rat. OECDP also show anti-inflammation and analgesia effects.
