Tartary buckwhea(tF . tataricum (L.) Gaertn.) with high pharmaceutical and edible values is a dicotyledonous plant.The bioactivity components in tartary buckwheat with antihypertension,antihyperglycemia and antihyperlipemia mainly is Flavonoids. In this paper a new process for purifying the active fraction from tartary buckwheat bran was investigated,and antioxidant activities and structure of refined tartary buckwheat flavonoids extraction from tartary buckwheat bran was studied.In order to select suitable macroporous resin as tartary buckwheat total flavonoids absorbent,static-absorption and static-desorption about fifteen kinds of macroporous resins which can purify flavonoids very well and the static-absorption dynamic curve four kinds of macroporous resins was studied.Adopted dynamic adsoption-desorption methods, the technological parameters of the purifying process of tartary buckwheat total flavonoids with the macroporous resin were optimized. By using superoxide radical system,hydroxyl radical system,DPPH system,H2O2 radical system and NO2ˉsystem, the antioxidation activities of tartary buckwheat total flavonoids extract were studied and compared with rutin and Vc. Its structure were identified by the spectral and Chromatography technique.The main contents of this paper:(1) Study on static-absorption and static-desorption about fifteen kinds of macroporous resins;(2) Study on the optimal dynamic adsoption-desorption technological parameters of the purification process of tartary buckwheat total flavonoids;(3) Study on antioxidant activities of refined tartary buckwheat total flavonoids extract;(4) Study on the structural identification of refined tartary buckwheat flavonoids extraction.The main results of this paper:(1) Static-absorption and static-desorption about fifteen kinds of macroporous resins which can purify flavonoids very well was studied. Through the comparisons among fifteen kinds of macroporous resin?s absorption rates, desorption rates and the static-absorption dynamic curve. The results showed that adsorptive quantity of total flavonoids is above 109 mg·g-1, adsorptive rate of total flavonoids adsorbed by DM-2 macroporous resin is above 83%, desorption rate of total flavonoids adsorbed by DM-2 macroporous resin is above 52%. The static-absorption dynamic curve is perfect. DM-2 macroporous resin is suitable absorbent to purify buckwheat flavonoids in the industry.(2) Response surface methodology(RSM)was employed in the study and a mathematical regression model for purification of tartary buckwheat total flavonoids with the macroporous resin was built. The technological parameters of the purifying process of tartary buckwheat total flavonoids with the macroporous resin were optimized. Results show that the optimal absorption parameters are adsorption velocity 2.0 mL·min-1, pH 2, solution concentration 2 mg·mL-1 and the optimal desorbtion parameters are 71.05% ethanol as eluting solvent, pH 8.76, the desorption velocity 1 mL·min-1. This method is simple and feasible with good effect of purification, which can meet industrial requirements.(3) By using superoxide radical system,hydroxyl radical system,DPPH system,H2O2 radical system and NO2ˉsystem, the antioxidation activities of tartary buckwheat total flavonoids extract were studied and compared with rutin and Vc. The results showed: tartary buckwheat total flavonoids extract has better scavenging effect in free radical system and the antioxidation activities of tartary buckwheat total flavonoids extract aren′t almost affected, and scavenging effect of some free radical is enhanced.(4) The flavonoids extracted from tartary buckwheat were purified by the macroporous resin.Its structure were identified by the color action,TLC,UV,HPLC. The results showed that structure of refined tartary buckwheat flavonoids extraction were determined as 5,7,3',4'-OH,3-O-rhal-6glu(rutin).The innovative points of this paper:(1) Suitable macroporous resin is selected as tartary buckwheat total flavonoids absorbent and response surface methodology was utilized to design experiment.(2) The antioxidation activities of refined tartary buckwheat total flavonoids extract were studied by different system.(3) Structure of refined tartary buckwheat flavonoids extraction was identified by the spectral and chromatography technique.
