Preparative chromatography is a better method with which we can get intermediates of drugs with high yield. The problems of the cost and the yield must be considered during the process of the preparative separation. So it is necessary to optimize the separation conditions. Preparative separation is usually performed under the nonlinear conditions and the objects to be separated are usually multiple, so it is important to recognize and analyze the parameters of the column system and to select the proper optimized process and method under such conditions. The paper is composed of two parts as follows.Firstly, based on the multiple components nonlinear chromatographic model and the two-component approximation, the isotherms of multi-component were fitted by inverse method with artificial compensation and then the relevant column parameters were measured in order to obtain EGCG from tea-polyphenols under preparative conditions.Secondly, the parameters for the nonlinear separation of EGCG were optimized. And in the optimum procedure the yield rate of EGCG was used as the objective function; the recovery yield and the purity of EGCG were used to be the constrained conditions.
