Xylooligosaccharides(XOS) that prominent capability as a prebiotic substrate for the proliferation of Bifidobacteria, a probiotic in the adult human colon, were received more and more attention with people more care about their healthy. The purification of XOS rooted in corn core, the calibration curves of different degree polymerization XOS and their fermentation by B. adolescentis were studied in this paper.The different purity of xylobiose(X2), xylotriose( X3), xyloteraose(X4), xylopentaose(X5), xylohexaose(X6), xyloheptaose(X7), and xylooctaose(X8) were harvested by ?KATAfplc separation flatpalm with the column XK50/60 filled with Bio-Gel P-2, with degassed ultro-purified water as eluent at 1.5ml/min at 48℃, 10ml XOS loaded, and detected by differential rafractometer. The purity of each saccharides were improved to 93.06%, 94.40%, 94.46%, 95.15%, 95.23%, 94.26%, 94.86% by column XK16/100 filled with Bio-Gel P-2, with degassed ultro-purified water as eluent at 0.2ml/min. The analysis of saccharides group indicated X4~X8 were composed by xylose and arabinose.The calibration equation of each saccharides were based on the relationship between concentration and peak height: X2 C=0.0002145*H+0.004337;X3 C=0.0001921*H+0.00226; X4 C=0.0002751*H-0.000353;X5 C=0.0002709*H-0.003423; X6 C=0.0002471*H+0.001294;X7 C=0.0002822*H+0.000293; X8 C=0.0002758*H-0.003140In the experiment of in vitro fermentation of XOS by B. adolescentis, it could concluded that the improvement of XOS'transform rate from17.85% to 20.44%, better consumption of saccharides'DP above 3, higher cell concentration could be realized by increased the concentration of X4~X7. It also showed the higher DP saccharides have better proliferation capability of B. adolescentis than lower DP. It was found that the cell concentration of B. adolescentis ascended slowly after exponential phase and didn't go to the death phase in the end of fermentation,when B. adolescentis grew in the tube that could vent metabolites into out environment. The proliferation of B. adolescentis by XOS came from degraded productions of xylan enzmetic hydrolysate residues was worse than enzymatic hydrolysate of XOS, but better...
