A lectin was isolated from the Rhizoma of polygonatum odoratum ( Mill.) Druce . The procedure involved aqueous extraction, (NH4)2SO4 precipitation, dialysis to remove (NH4)2S04, ion exchange chromatography on CM-Sepharose, gel filtration on S-200. The lectin content was about 40 mg/100g dry flour . The lectin can agglutinate rabbit, and can not agglutinate human blood cells. Mannosans was the specific inhibitor of the POL hemagglutinating activity. The native molecular mass of the lectin detected by S-100 was 48kDa. And when examined by SDS-PAGE , this lectin gave a single band of 14. 2kDa in the presence of 2-mercaptoethanol. The lectin is stable up to 60°C and is stable in pH3-9.The fluorescence spectrum (FLS) of POL excited at 280nm and 295nm showed a maximum peak at 328nm. The characteristic peak of Tyr did not exist, and it showed that the fluorescence energy of Tyr was transformed to Trp and strength the fluorescence of Trp. When POL was excited at 295nm, the FLS showed a maximum peak at 328nm, the λ? of fluorescence emission...
