Rhizom Corydalis is one of Chinese traditional drugs with the function of promoting flow of Qi and Blood and of stopping pain. As a drug, it can be traced back to Kai Bao Ben Cao and it is dry stem tuber of yanhusuo W.T.Wang. Modern pharmacology research shows that Rhizom Corydalis has the functions of easing pain, lulling, resisting the ulcer, suppressing the gastric acid to secrete, solving the convulsion and increasing the flow of coronary artery blood etc., and has extensive application on clinic. The base of traditional Chinese medicine (TCM) researching and developing is traditional Chinese medicinal material, of which quality control is the key of modernization of TCM. Pharmacokinetics research of TCM is of important significance for optimizing the preparation craft, making the standard of quality control, realizing the TCM modernization and moving towards to the international market. So, this subject studied quality and pharmacokinetics of Rhizom Corydalis.There are two parts in this thesis:Part one: Review1. Study survey of Rhizom CorydalisIt includes Chemical composition summary, pharmacology research and pharmacokinetics study progress. The backbone of Rhizom Corydalis is the alkaloid, what chemical composition studied most is the alkaloid too. More than 40 kinds of chemical structures of Rhizom Corydalis alkaloid have already been appraised. The drug has extensive pharmacology activation, and the pharmacology research about it involves central nervous system, cardiovascular system, digestive system, hypophysis-renal gland cortex system etc. The study progress part of pharmacokinetics consists of research approach of TCM and effective composition studies of Rhizom Corydalis.2. Progress in the quality research of Rhizom CorydalisIt involves application and progress of fingerprint in the quality control of TCM, current situation of quality evaluation and the question in quality evaluation of Rhizom Corydalis.Part Two: Research papersIn this part, four departments are involved.First, Study on Ultrasonic Extraction of total Alkaloids from Rhizoma Corydalis Sonochemistry, as a new branch of subject, can quicken chemical reaction and enhance productivity. The total alkaloid was extracted with 70% ethanol under the action of ultra-sound, in which the contents of tetrahydropalmatine and protopine were determined with high-performance liquid chromatographic method (HPLC). Such factors about the extraction as the ultrasonic time, temperature, power and solvent amount, acidity and concentration were investigated. This method offers the theoretical foundation and craft parameter for industrialized extraction of the total alkaloids.Second, Simultaneous determination of tetrahydropalmatine and protopine in Rhizoma CorydalisThis experiment established a reserved-phase HPLC method for simultaneous determination the contents of tetrahydropalmatine and protopine in Rhizoma Corydalis from different habitats. The average recoveries for tetrahydropalmatine and protopine were 101.67% and 98.13%, and R.S.D. were 1.44% and 1.37% at wavelength of 281nm, respectively. The method offers the scientific method for two alkaloids of Rhizoma Corydalis to be determined at the same time and established the foundation for quality evaluation of the drug.Third, Establishment of chromatographic fingerprint and quality assessment for Rhizoma CorydalisThe chromatogram separation was carried out using the gradient elute method. Data processing to compare and analyze the similar degree of fingerprint of the drug from different habitats and with different process method was carried out with "chromatogram fingerprint of TCM similar degree appraise" software (2004A edition) prepared by northwest university. At the same time cluster's analysis to 21 batches of samples was carried out utilizing fingerprint of the contrasting drug.
