Most of pharmaceutical molecular have a larger conjugated structure. Some time their oxidative product would produce fluorescence or have greater fluorescence intension. Some colorless molecular could also be measured by spectrometric method when they were oxidized.By the integration reagent immobilization with flow injection technique, the new methods for determination of folic acid and Chlorpromazine hydrochloride have been established in this thesis, respectively, in which the immobilized phosphate ferric and dioxide lead were used as oxidant. The interaction between Chlorpromazine hydrochloride and protein was also studied.Part one: reviewIn this part, the new technique and methods of inorganic reagent immobilization in recent years were discussed. Four immobilized processes and their application in flow injection analysis were reviewed in detail.Part two: Study reportsIn this part, two analytical systems have been investigated.1. Fluorescent analysis study for folic acid with a flow injection solid-phase reactor system(1) Flow-injection fluorescent determination of folic acid in tabletA dioxide lead solid-phase reactor was constructed using polymeric embed method. Based on the greater fluorescence intensity of oxidized product, a new method of flow injection fluorescent determination for folic acid in tablet was established with solid-phase reactor on-line oxidation. In phosphoric buffer medium and a 80 ℃ water bath with a 15cm solid-phase reactor, the provided method has a wider line range(8×10-9~2.5×10-6 g/mL) and highly sensitivity(1.0×10-10g/mL), the RSD of 11 replicate determinations is less than 0.85%. The method has been applied to determine the folic acid in tablet and the results were basically agreed with standard method.(2) Fluorescent determination of dissolution rate for folic acid tablet with a flow injection on-line filtration and dilution systemThis method was based on the previous one and has a relatively good results comparing to standard method. It was proved that the dissolution rate of tablet was over 75 percentage in 45 minutes.(3) Flow injection fluorescent determination of folic acid in human blood with a dialysis separator systemIn order to avoid the fluorescence emitted by protein in blood, a home-made dialysis separator has been used. The recoveries were investigated in there people ranging from 95.5 to 108%.2. Fluorescent analysis study for Chlorpromazine hydrochloride based on flow injection on-line oxidation with solid-phase reactor(1) Flow-injection fluorescent determination of Chlorpromazine hydrochloride Chlorpromazine hydrochloride would have greater fluorescence intensity when itis oxidized. A new method for fluorescent determination of Chlorpromazine hydrochloride was presented based on flow injection on-line oxidation with solid-phase reactor. A 5cm of dioxide lead oxidant column was used as solid-phase reactor. The line range is 1×10~8~4X 10"6 g/mL, and a detection limit of 1.6×10~9g /mL was achieved, the RSD of 11 replicate determinations is less than 1.6%. 60 samples can be analyzed per hour. The proposal method was successfully applied to the determination of Chlorpromazine hydrochloride content in various pharmaceutical preparations, and the results were agreed with official method.(2) Fluorescent determination of dissolution rate of Chlorpromazine hydrochloride with a flow injection on-line filtration and dilution systemThe process of this method is as same as the previous and the results obtained were statistically compared with official method. The dissolution rate was over about 70% within 44min.(3) Study on the interaction of Chlorpromazine hydrochloride with protein by flow-injection solid-phase reactor fluorometry with a dialysis separation systemA flow-injection solid-phase reactor fluorometry combined with dialysis separation for studying protein binding of Chlorpromazine hydrochloride was carried out.
