In this thesis, we designed and synthesized a series of dualistic and triple complexes based on the complexe of chitosan and zinc. They were characterized and studied with kinetics,urea-adsorption,difference ability of coordination among diversified molecular-weight chitosan,etc. In addition,a new chemiluminescence method for the determination of amino acid with reverse flow injection technique was developed. Some satisfying results were obtained which are theoretic base for the further applications. The thesis was divided into six parts:Part â… : Review.A review of the complexes of chitosan and its derivatives and the application of chitosan and its derivatives on medicine and biology. Part â…¡ : A kinetic study on the complex of Zn(â…¡)-chitosan.The adsorption effect of zinc ions on chitosan was investigated by adsorption method.The study of the adsorption kinetics showed that the rate of adsorption effect of zinc ion on chitosan can be interpreted in term of first order reaction .The experimental data of adsorption equilibrium from zinc ion solutions were satisfied with the Langmuir and Fruendlich isotherm equation.The composition and structure of chitosan-Zn(â…¡) complex were investigated by element analysis, UV-visible spectrum and IR spectrum.The results show that the ratio of Zn(â…¡) with chitosan was 3:1,and the possible composition of the complex was [Zn(CTS)s]. SO4.6H2O.The non-isothermal kinetics thermal parameter were obtained form the analysis of TG-DSC curve by differential and integral methods.The most possible mechanism function,the activation energy,E and the pre-exponential constant,A was obtained.Part â…¢: Sorption of urea by complex of chitosan-Zn(â…¡). A method for adsorption of urea with chitosan-Zn(â…¡) complex was studied. The optimal condition was obtained from experiments.The adsorption capacity of the complex for urea was83.97mg/g under room temperature,pH value between 6-7,the original concentration of urea was 2.0mg/mL.The adsorption time was 3.0h.Part â…£: The study of different chitosan degradation complexes with zinc. Chitosan was degraded by H2O2 and a series of different molecular weight chitosan was obtained.The adsorption behavior of chitosan on Zn(â…¡) was studied.The structure of the complexes was obtained by element analysis,Uv-vis spectrum and IR spectrum.The adsorption capacity of the complexes of CTS-Zn(â…¡) on urea was also studied.Part V : Synthesis and characterization of the complex of chitosan-zinc-N-salicylidene glycine schiff base and the effect on the elimination of superoxide anion. Chitosan-znic-N-salicylidene glycine schiff base has been synthesized relying on the coordinative potential of chitosan and characterized by element analysis,IR spectrum,UV-Vis spectrum,and thermal analysis .The effect of this complex on the elimination of superoxide anion was investigated by Pyrogallol-NBT colorimetry. Part VI : Determination of amino acid by reverse flow-injection chemiluminescence. A new chemiluminescence method for the determination of amino acid with reverse flow injection technique was developed under the alkaline condition.The method is based on the chemiluminescence reaction between ferricyanide and Luminol,for amino acid can increase the chemiluminescence intensity. The detection limit can be 0.01ug/mL,sampling frequency is 180h-1.The relative standard deviation is 1.21%(4ug/mL L-arginine,n=12).The rate of recovery is between 98.2-102.4%.The separately linear range for tyrosine,glycine and arginine is 0.1-40ug/mL, 0.01-1.0ug/mL, 0.01-1.0ug/mL.
