Fish oil is a kind of natural functional food and its availability is high. Its main active components are PUFA such as EPA and DHA. At present the development of functional food offish oil in our country is short. The method of extraction was organic solvent, and the quality of the product was not very good. In this study supercritical CO: extraction was used for extraction of EPA and DHA in fish oil. The parameters in supercritical CO2 extraction and separation were studied. The difference between supercritical CO2 extraction and other extraction methods was showed.1 Esterification experiment showed that the best method was quick esterification whose process was simple and the reagents used were common.2 The results of Urea inclusioncomplex showed that urea could crystallize with saturated fatty acid and low unsaturated fatty acid (LUFA), thus EPA and DHA were extracted. And lower temperature was available to form the product of high concentration.3 PUFA could be enriched by AgNO3 -complex strongly and specially, but the method was limited by the composition of the product.4 The parameters in supercritical CC>2 extraction and separation were studied. Revolving, orthogonal and tropical design of the second power of three factors was adopted to choose the optimum technological conditions of supercritical CO2 extraction which were extraction temperature of 40 癈, pressure of 26MPa and time of 114min. Temperature gradient of rectification was 45℃, 50℃, 55℃, 70℃.5 The products of supercritical CO2 extraction, urea inclusioncomplex and AgNO3-complex were compared. The result showed that supercritical CO2 extraction was better than other two methods.6 The experiment of antioxidation showed that the effect of TBHQ was the best when only one antioxidant was added. When four antioxidants were added altogether the formula was 0.02% of TBHQ, 0.01% of BHA, 0.01% of VE and 0.005% of PG.
