Studies On Extraction & Purification Processes Of Total Saponins Of Ilex Kudingcha

Ilex Kudingcha C.J Tseng is widely used as a traditional beverage in southern China, It is also used in popular medicines and in commercial herbal preparations as a stimulant to the central nervous system,an aid to losing weight and for the relief of hypertension.lt mainly contains triterpenoid saponins and other components such as sterols, amino acid ?vitamins ?lipopolysaccharide and many microelements . In-abroad ,the research about Ilex Kudingcha is concentrated on chemical components and pharmacology ,and there is few reports on the extraction and purification.On the bases of previous research into Ilex Kudingcha ,and in consideration of specific resources of the laboratory .This article optimized the parameters of ultrasonic extraction and the parameters of reflux extraction which was screened by the orthogonal design.The purifacation prosess of total saponins with macroporous resin was generally studied by taking adsorption ratio ,eluting ratio and purify of total saponins as indices.The results were as follow:1.The optimal parameters of the ultrasonic extraction were that : dipped in the 70%EtOH(20 folds) for lh,then extracted for 0.5h by ultrasonic,20MHz.2.The optimal parameters of the reflux extraction were that:dipped in the70%EtOH (12 folds) for Ih, then refluxed for 0.5h ,3 times altogether.3.The study of purification process showed that D101Bmacroresin had themaximum adsorption ratio among three resins:D101A,D101B and AB-8. Its dynamic and static state adsorption ratio were : 92.0+0.7mg.g-l dried resin, 116.2+1.9mg.g-l dried resin; the elution ratio was 93.13+0.21%. The experiments showed the optimal parameters of purification were that:original concentration(10-14nig/ml) ,PH value (6.0-7.0), velocity of flow (2BV/h), then eluted by water(4BV) ,30%EtOH(4BV), 70%EtOH(4BV), gather the part eluted by 70%EtOH, and total saponinss in the solid extract could reach 61.82+0.32%, RSD:0.52%, the refine ratio could reach at 247.44%.The results of the whole experiments had provided basic data for preparation process and quality control of modern Chinese Pharmaceuticals intend for the treatment of hypertension,which was a reference for process improvement and quality control of similar Chinese herbal Pharmaceuticals as well.In the study ,the application of ultrasonic extract and macroporous resin primarily demonstrated that these new techniques and new materials could enhance extrating and purifying ratios of effective components in Chinese herbs,and they could be feasible to promote their application to Chinese herbal pharmaceuticals R&D.
