Initial the glioma-amplified sequence (GAS)41 gene was identified in the glioblastoma multiforme cell line TX3868.The gene was found to be frequently amplified in glioblastomas and astrocytomas and thought to be involved in early gliomal tumor development. GAS41 is highly conserved in evolution, and it encodes a protein that is highly conserved among human,mouse,chicken,zebrafish,Drosophila melanogaster,yeast and Caenorhabditis elegans. GAS41 has an N-terminal YEATS domain,followed by an A box and B box at the C-terminus. GAS41 is a member of the YEATS domain family. The YEATS domain family of members, conserved from yeast to human more than 70 eukaryotic species.It is evident that the YEATS domain proteins have some function in chromatin modification and transcription,but the specific function of YEATS proteins in this process is not clear.Although GAS41 is essential for cell growth and viability, until recently, the research on GAS41's function focused on the interaction with other transcription factors activate the transcription process,but for the research of the GAS41's function of the development of organisms is less.In this study,we use the zebrafish as a model organism,cloning of the zebrafish gene of GAS41,by RT-PCR, in situ hybridization, Morpholino antisense oligos technology,over-expression analysis tools such as experimental study of GAS41 in zebrafish embryos during the early development of the functions., we found that GAS41 knockdown or over-express will have effect on head development., GAS41 may play an important role on the development of head in early embryonic development.
