Water eutrophication is most prominent of globale environmental prolem.Although microbes are mostly sensitive and fastly responded to environmental changes,the relationships between microbes and water eutrophiccation is still in argument now. Therefore the paper studied how related between prokaryotic microbes and environment varibles especially N and P,which are normally the source of water eutrophicated,in hypereutrophicated Lake Wuliangsuhai,to rich and development the correlative theroy, and to provide the bases for water environment protection and remedy.Wuliangsuhai Lake is located in semi-desert areas of the world,which is extremely rare with high eco-efficiency of large-scale multi-purpose lake.Their special ecological environment will inevitably contain a large number of function-specific micro-organisms. At present,Wuliangsuhai Lake is polluted,so water quality trends towards extreme eutrophication,the ecological environment has become extreme,the study on also biodiversity of the lake is very important.In particular,no studies have reported diversity of prokaryotic microorganisms in Lake Wuliangsuhai.The water and sediment samples used in this study were collected from the HGB,SJB and XK sites in the Wuliangsuhai Lake.HGB site is located in the central zone of the lake,which is the largest concentration of nutrients;XK site is located in the southern part of the lake,which is the smaller concentration of nutrients;SJB site islocated in the eastern part of the lake.In this study,total DNA were extracted directly from water and sediments in Wuliangsuhai Lake,using as template for for polymerase chain reaction(PCR) amplification,which employ universal primer for bacteria and archaea 16SrRNA gene seqnences.The 16SrRNA gene libraries were then constructed.This paper investigated and analyzed the diversity and phylogeny of bacteria from water and archaea from sediments in Wuliangsuhai Lake,by using the construction of 16S rRNA gene clone library.And,to assess the microbial diversity through calculation and analysis of phylotype diversity index and the correlation coefficient.Furthermore,to analysis the relationship between the extent of eutrophication and the structure of bacteria.The results showed that the results showed that most of which were affiliated with bacterial divisions commonly found in freshwater ecosystem,e.g.α-,β-,γ- proteobacteria, cytophaga-flavobacter-bacteroidetes,Actinobacteria.β-proteobacteria and CFB were dominant linage in Lake Wuliangsuhai.there are distinct differences in the microbial community structure and diversity of different sampling sites,which is influenced with the degree of eutrophication changes.In Archaea domain,it was found Crenarchaeota and Euryarchaeota kingdom and predominate by Crenarchaeota.There are distinct differences in the microbial community structure at different depths of sediments.The analyses indicated the relatively high diversity of bacteria in this sediment environment and the shift of bacterial communities with depth should also indicate changes of physichemical properties on fine scale of sediments close to surface.Comparison of different nutritional analysis showed that bacterial community structures from three sites are different in Lake Wuliangsuhai,and with increase of the degree of eutrophication,microbial diversity is a lower level.It shows that the structure of Community structuresmay is related to lake eutrophication.
