Zds1/Sch9 Function In Regulating The Ras-cAMP Signaling Pathway In The Yeast Saccharomyces Cerevisiae

The function of Sch9/Zds1 plays important roles in regulating the PKA activity so as to heat shock resistance and the localization of its regulating subunit Bcy1 in the Budding Yeast Saccharomyces Cerevisiae.Meanwhile,the localization of Bcy1 also has close relationship with carbon source dependent manner. Investigating the roles of Sch9/Zds1 together with the carbon source dependent manner in regulating the activity of PKA and the localization of Bcy1,will not only help to intensify our current research of signaling pathway of Budding Yeast Saccharomyces Cerevisiae,but also offer references to other higher eukaryotes.Our work indicated that overexpression of ZDS1 in the strain with normal PKA level lowered its PKA level,though overexpression of ZDS1 in the strain with low PKA phenotype led to the increase of PKA level.These result illustrated that Zds1 may regulate PKA level in response to different PKA phenotype by itself in the yeast.in contrast with the wild type,Bcy1 in the zds1Δmutant and sch9Δmutant all imported to the nucleus under different carbon sources. Overexpression of ZDS1 in the wild type and the sch9Δmutant led to the cytoplasmic accumulation of Bcy1. overexpression of SCH9 in the wild type also resulted in the cytoplasmic accumulation of Bcy1, whereas, the same treat with the zds1Δmutant can't bring on the cytoplasmic accumulation of Bcy1.Through these results,we can surmise that SCH9 is likely to locate in the upstream of ZDS1 regulating the localization of Bcy1.Compared the results of heat shock assay with the ones of fluorescent assay,we found that the two parts of results have same trend,which indicate that the activity of PKA may has direct relationship with the localization of Bcy1.We also find that in part of the cells which are in the reproduction of budding, Zds1 accumulated in the bud neck, from which we can infer that Zds1 may play an significant role in the process of reproduction of budding in the Budding Yeast Saccharomyces Cerevisiae.
