Artemisia sphaerocephala Krasch(ASK) is of great amount in our country. The extraction rate of its seeds polysaccharides is about 7%. It has HyPoglycemic Effect, et.al.. However, there's little research conducted on the physico-chemical properties, structure and biological activities of water soluble Artemisia sphaerocephala Krasch Seeds polysaccharides (ASP). In this paper, ASK seeds was studied by the advanced methods of isolation and purification technique, chemical and instrumental analysis technique of bio- macromolecular .The main results are as follows:1. The extraction and physico-chemical properties of crude water soluble ASPCrude water soluble ASP was obtained from ASK seeds by water and precipitated with ethanol. Gas chromatography analysis showed that the polysaccharide was composed with Rha, Fuc, Ara, Xyl, Man, Glc, Gal, GlcA and GalA, their molar ratio was 13.49:1:61.83:26.37:3.49:6.70:66.76:6.68:29.81. The total sugar content is 95.65 %, uronic acid content is 36.92 %. The UV spectrum shows there is no protein in this polysaccharide, and the IR spectrum shows polysaccharides feature absorption.2. The isolation, purification of water soluble ASP and physico-chemical properties of its homogeneous component ASP-AASP-1, ASP-2, ASP-3, ASP-4 was isolated by DEAE-Cellulose column. From ASP-1, ASP-A was purified by Bio-Gel P-150 column, proved to be homogeneous on molecular weight by HPGPC, and its Molecular Weight is 55.1 kDa. Gas chromatography analysis showed that the polysaccharide was composed with Ara, Man, Glc, Gal, their molar ratio was 1:5.00:9.21:3.39.3. Studies on the structure of ASP-AThe IR spectrum shows furanose feature absorption. Based on the methylation analysis by GC-MS, there are mainly 2,3,5-Me3-Ara, 2,4-Me2-Ara, 2,3,4,6-Me4-Man, 2,3,4,6-Me4-Gal, 2,3,6-Me3-Glc and 2,3-Me2-Glc in the methylation ASP-A. Inferred from analysis of partial hydrolysis, the core structure of polysaccharide's main chain of ASP-A are mainly made up of Man and Glc, the side chain and the non-reduced end are composed of Ara and Gal.4. Studies on the functions of crude water soluble ASP and its component ASP-A when gives to immune cells of miceThe results showed that ASP-A strongly stimulated proliferation of mice spleen lymphocyte when used with Con A. However, crude water soluble ASP didn't have any functions about this.
