The Pine nut Shell Polysaccharide (PSP) is one kind of polysaccharide ingredient which husked from the pine shell, The research demonstrated that PSP plays a important role in anti-tumor, anti-virus,etc. However, most studies of PSP stand the situation of direct anti-virus and anti-tumor and few studies on immune cells and cytokines were reported by consulting literature. In order to obtain the influence on immune cells and cytokines by PSP, cytotoxicity on mouse splenic lymphocytes, proliferation of T and B lymphocytes induced by ConA or LPS were determined by using MTT assay. Peritoneal macrophage function was determined by using neutral red approach. NK cell competence was determined by using lactate dehydrogenase approach. Contents of IL-2 secreted by T splenic lymphocytes , content of TNF-αsecreted by peritoneal macrophages was determined by using DAS-ELISA approach.The results showed that PSP cell toxicity to splenic lymphocytes is very low, It remained higher immunologic competence on splenic lymphocytes Even if the polysaccharide density achieves 1000μg/mL.PSP of different concentrations can promote T and B lymphocyte proliferation,and PSP significantly enhanced T lymphocyte proliferation induced by ConA ranging from 100μg/mL to 200μg/mL (P<0.01). Different density PSP can enhance the function of peritoneal macrophage englobing neutral red and its metabolism. PSP can improve effect of NK cells against YAC-1. Different density PSP can promote IL-2 secretion by splenic lymphocytes plus ConA.PSP significantly promote IL-2 secretion by splenic lymphocytes plus ConA ranging from 50μg/mL to 200μg/mL (P<0.01). TNF-αsecretion was significantly improved by PSP of different concentrations, In which 100μg/mL and 200μg/mL group presses secretion quite emarkable(P<0.05). PSP can strengthens immune cell immunity activeness from immune cell level and cytokines level,and PSP can improve induction of related anti-tumor cytokines. The experimental result prompt, PSP very strongly immunity activeness and has a low toxic characteristic. The Pine nut Shell Polysaccharide seems to be a good didate for the development of Immunity conditioner.
