The thesis unites theory with practice and studies the technique of extractingRNA with beer yeast as the raw material through all kinds of contrast trails andstability experiments. This technique is highly effective and practical according tothe statistics and the targets shown by the trails. It can effectively extract RNAwith relatively high value from the waste beer yeast. It obtains the considerableeconomic benefit while realizing the recycling waste material, reducing pollutionand protecting the environment.The thesis respectively discusses the recovery, the comprehensive utilizationand the wide use of RNA. The principle of RNA extraction, the technique, theprocedure, the choice of the most suitable condition, the extraction, the stabilitytests and the determination methods of RNA are also discussed more explicitly.The prospects of the frontal technique of extraction are explained briefly.This question for discussion studies the comprehensive utilization of thewaste beer yeast and intends to adopt the new technique to extract the RNA fromthe waste beer yeast. The concrete experiment is to take the waste beer yeast ofKinsberg Beer Company as the raw material, adopt the watery salt method toextract RNA and explains the principle of extraction and the choice of the mostsuitable condition. The condition contrast tests are made for the temperature ofextraction, Ph value, the amount of salt added, the time and the concentration inorder to confirm the optimum value. The result of the experiment shows that thistechnique of extraction RNA is relatively stable. The average rate of achievementcan reach 3.32%, the purity of the RNA can reach 87.8% and the rate of therecovery of de-nucleus yeast can reach 65.4%. All reach the targets that the plansfix. This technique is simple and practical. It can be easily operated andmaintained. The investment is small, the benefit is good and it's economicallypractical.During the trials, the question for discussion also tries the new technique ofthe RNA extraction with the low salt (6%), the neo-aeroflocs and the depositaccelerator under the normal temperature. Different from the technique of thethick salt and the low temperature, this technique makes the purity of RNA meetup to the standards, the yield is high. It can also save the refrigerator installation,shorten the time of the RNA deposit and contribute to the conservation of thede-nucleus yeast.Besides, the test also adopts the aeroflocs of crust elements to take place ofthe one of polyacrylamide to accelerate the deposit of the protein in order to purethe RNA. It's very effective and also avoid the second-time pollution. In brief,adopting our new technique and the best condition fixed through a large numberof experiments can effectively extract the RNA with the relatively high valuefrom the waste beer yeast. It can realize the recycling waste material, reducingpollution and protecting the environment.Finally, the thesis also tentatively studies the technique of how to usenanometer equipment to smash the yeast cells in order to extract RNA.
