Spermatogonial stem cells were located at seminiferous tubule which not only can maintain the number of themselves but also can differentiate to spermatocyte. Because of its potential at transgenc animals production , it had became a hotspot in the research of stem cell. The study was performed to characterize effects of three media on culture of chicken SSCs in vitro when there is layer or not and to select the best culture medium, investigate characteration of SSCs and the proper conditions and factors when SSCs were subcultured, compare the effect of exogenous cell factors as mLIF, hSCF and bFGF on SSCs proliferation and differentiation and discuss the control mechanism of those cell factors. We attempted to prepare the ground for the establishment of chicken SSCs lines and offer cell model for growth and differentiation of cells.The main results were as follows:1.SSCs after they were isolated and purified by Percoll density centrifugation were cultured in six culture medium. We observed the growth character of SSCs and compared the rate of SSCs colony, the results showed: Although SSCs can survive in culture medium I, IIand III without layer cells, the longevity was short and no colony was observed.
