The bioinformation of international data base was explored in the present research. Nucleotide sequences of rDNA ITS region from 90 grass speiece,23 genera were obtained from Genbank database of NCBI by using EntrenZ and Blastn similarity searching functions. The objectives of present research were to construct a phylogenetic tree with ITS mucleotide sequences in order to provide additional information on the evolution and classification of herbaceous plants in grass family and to validate the feasibility of the approach in plant phylogenetic study.Complete sequences of rDNA ITS region include ITS1,5.8S and ITS2. They were obtained by searching of Genbank of NCBI and related websites. The sequence names from Blastn and Entrez were used as keywords to search for homologous sequences.Selected sequences were output from NCBI sequence viewer, saved by NCBI format with NCBI locus names. After being converted by using DNAStar Bioedit and certain DOS command,the data of 90 species from 23 genera imported into ClustalX for alignment, Homologous comparison and statistical analysis.The aligned sequences data were input into Mega 3.0 and various phylogenetic trees were constructed by using Mega 3.0 and Phylips programs.Our results indicated that the classification by ITS sequences was basically same as conventional classification. The evolutionary sequence of the plants could be determined by relative lengths of tree branches for each species. Some points needing attention were also discussed while exploring international database,e.g., Genbank.
