| Objective:Based on the data mining technology,the rule of Wang Tan’s treatment of non-small cell lung cancer was analyzed to provide clinical reference.Through network pharmacology and subcutaneous tumor transplantation model of tumorbearing mice,the inhibitory effect and potential molecular mechanism of the representative prescription of warming liver and nourishing spirit(Chu Ji Xiao Yi decoction)on non-small cell lung cancer were investigated.Methods:1.The clinical study collected non-small cell lung cancer patients who visited Professor Wang Tan’s outpatient clinic from October 2019 to September 2022.Case information includes basic information,clinical symptoms,auxiliary examinations,TCM diagnosis,TCM syndrome types,Western medicine diagnosis and pathological data,TCM prescriptions,etc.Establish a patient database,and use the TCM inheritance computing platform V3.0 to conduct statistical analysis of medicines,odors,meridians,efficacy,etc.,and determine core prescriptions.2.Preliminary exploration of the mechanism of core prescription treatment of non-small cell lung cancer through network pharmacology.3.A mouse model of xenograft tumor was established and randomized.Intragastric intervention: The traditional Chinese medicine group was given 0.2ml of the corresponding concentration of liquid medicine(3.64g/kg,7.28g/kg,14.56g/kg);The control group and the cisplatin group were given 0.2ml of normal saline by gavage.Injection intervention:cisplatin group cisplatin(drug dose of 3mg/kg)intraperitoneal injection,1 injection every 3days;The rest of the group were intraperitoneally injected with normal saline.After 21 days of intervention,tumor weight was observed,and the tumor inhibition rate,HE staining of lung cancer tissues,immunohistochemistry detection of lung cancer tissue proliferation,TUNEL method detection of lung cancer tissue apoptosis,Western blot detection of tumor LC3 II.,Beclin-1 expression and activation of PI3K/AKT/mTOR pathway were calculated.Results:Through data mining of 341 cases of prescription in clinical study,35 high frequency drugs were obtained.Through cluster analysis and association analysis,the core prescription was determined to be Chu Ji Xiao Yi decoction(dried ginger,Baizhu,ginseng,scorched licorice,trichosanthin,Paeonia alba,coix seed,ephedra,frankincense,myrrsula,Yizhi kernel,turtle shell,turtle shell,jujube kernel,poria cocos,cooked ground,althea bark).Three prescription groups were obtained by analyzing high-frequency addition and subtraction drugs.After analysis,the results of this study are consistent with Professor Wang Tan’s clinical use of warm liver and spirit treatment of non-small cell lung cancer.2.Network pharmacology predicts that the inhibition of tumor by eliminating Chu Ji Xiao Yi decoction may be related to the intervention of PI3K/AKT/mTOR signaling pathway.3.The experimental study showed that Chu Ji Xiao Yi decoction decoction could reduce the tumor quality of tumor-bearing mice;In terms of tumor inhibition rate,the low dose,medium dose and high dose groups were 10%,23% and 44%,respectively.The necrosis area of tumor tissue in tumor-bearing mice was increased,the proliferation of tumor cells was inhibited,and the apoptosis of tumor tissue cells was promoted.The expression levels of LC3-II and Beclin-1 in lung cancer tissue of tumorbearing mice were significantly increased after treatment with Chu Ji Xiao Yi decoction decoction.The expression of PI3K/AKT/mTOR signaling pathway can be inhibited by eliminating the accumulation and eliminating dissolving.Conclusion:Clinical research: Professor Wang Tan believes that the main pathogenesis of non-small cell lung cancer is Yang deficiency and liver stagnation,and the mind and spirit are not used.In treatment,warm liver and spirit cultivation are advocated.Through data mining,the representative prescription of warm liver and spirit cultivation method is summarized-Chu Ji Xiao Yi decoction.Experimental study: Chu Ji Xiao Yi decoction can promote the autophagy of lung cancer cells,inhibit the proliferation of lung cancer cells,promote the apoptosis of lung cancer cells,so as to achieve the effect of tumor inhibition.The mechanism may be related to the inhibition of PI3K/AKT/mTOR signaling pathway. |
