Objective: A study on the mechanism of Huoxuetongdu Decoction for spinal cord injury based on metabolomics and lipidomics.Methods:The spinal cord injury model of rats was established by Allen’s percussion method.After the operation,the rats were administered with Blood Activating Tonics and saline by gavage for different groups,and the activity of the rats was continuously observed and recorded during the period.7 days later,the spinal cords were taken and morphological observations such as paraffin sections and Neu N staining of nerve cells were performed.Results:1.Behavioral scores of experimental animals: sensory and motor functions were poorer in the model group and the Huoxuetongdu Decoction group than in the sham-operated group after spinal cord injury(P<0.05),compared with the model group,starting from about the third day after surgery.The recovery of sensory and motor functions of the rats with spinal cord injury in the Huoxuetongdu Decoction group was better than that of the rats with spinal cord injury in the model group(P<0.05 or P<0.01);2.morphological observation of experimental animals: the spinal cord of the shamoperated group was structurally intact,with clear gray-white matter boundaries in the shape of butterflies,and obvious clear tiger-like Nisus bodies were visible in the nerve cells;compared with the model group,the spinal cord stasis foci were small,the swelling of nerve cells was lighter,and the morphology of neurons was better in the Huoxuetongdu Decoction group,and no obvious vacuole-like changes were seen;3.applied a metabolomics method based on UHPLC-QTRAP MS technology,which was used to conduct a metabolomics study of the effects of Huoxuetongdu Decoction on spinal cord injury repair and elucidate the related metabolic pathways.4.applied a lipidomics approach based on UHPLC-QTRAP MS technology,which was used to conduct a lipidomics study of the effect of Huoxuetongdu Decoction on the repair of spinal cord injury Conclusion:1.Huoxuetongdu Decoction can promote the recovery of sensory and motor functions of the hind limbs in the early stage of acute spinal cord injury;2.Huoxuetongdu Decoction can reduce apoptosis by promoting Neu N and inhibiting the expression of Caspase-3,thus facilitating the repair of acute spinal cord injury.3.Huoxuetongdu Decoction can interact with multiple metabolic pathways so as to repair spinal cord injury diseases. |
