| Backgroud: circular RNA(circRNA),as a kind of non-coding RNA,is involved in regulating various biological functions during the development of cancer.However,the function of circRNA circUBE2J2 in hepatocellular carcinoma is largely unknown.Methods: We performed Circ RNA micro Array to identify circRNAs that are aberrantly expressed in hepatocellular carcinoma and adjacent normal tissues.QRT-PCR was used to detect the expression levels of various RNAs.Kaplan-Meier survival analysis was used to explore the significance of circUBE2J2(hsa_circ_0009246)in clinical prognosis.Then,we explored the functions of circUBE2J2 in HCC by CCK-8 assay,plate clone formation,Ed U infiltration assay,Transwell cell migration and invasion,flow cytometry cycle analysis and mouse xenograft experiments.Dual-luciferase reporter gene transcription analysis and RNA-binding protein immunoprecipitation experiments to verify the relationship between circUBE2J2,miRNA-370-5P and KLF7.Results: circUBE2J2 was downregulated in HCC.Kaplan Meier survival analysis showed that downregulated circUBE2J2 was correlated with poorer survival.Knockdown of circUBE2J2 in HCC could downregulate KLF7,the target of miR-370-5P,resulting in the promotion of HCC cell proliferation and migration,while overexpression of circUBE2J2 could inhibit the proliferation and migration of HCC cells.Subsequent experiments showed that circUBE2J2 and KLF7 reulated the expression of each other by sponging miR-370-5P.Conclusions: Taken together,we conclude that circUBE2J2 may function as a competing endogenous RNA(Competitive endogenous RNAs,ce RNA)to regulate KLF7 expression through sponging miR-370-5P and play a regulatory functions in HCC. |
