than 50 years old.Factors such as diet,lifestyle and sponge interactions between long-stranded non-coding RNA(lncRNA)and microRNA(miRNA)play an important role in PCa.Studies have shown that dietary composition or structure is one of the suspected risk factors for prostate cancer,and abnormal long non-coding chromatin-associated RNA10(lncCAR10)and microRNA-203(miR-203)can promote the biological process of tumors,however,lncCAR10 and miR-203 on the biological behavior of prostate cancer and its mechanism,correlation studies have not been related The correlation study has not been reported.The aim of this study is to investigate the relationship between diet and lifestyle factors and prostate malignancy from both epidemiological and genetic aspects,and the role and mechanism of lncCAR10/miR-203/RSG17 axis in the biological behavior of prostate cancer,with the aim of providing theoretical guidance for tertiary prevention of prostate cancer and providing sensitive and specific relationship in prostate cancer A retrospective analysis of the clinical data related to a total of 283 patients admitted for inpatient treatment of prostate biopsy was performed,and the patients were divided into prostate cancer group and non-prostate cancer group according to the puncture results.The general characteristics of the two groups were compared and risk factors related to diet and lifestyle of patients who Underwent prostate puncture were analysed.Part 2:Exploration of the effect of miR-203/RGS17 on the biological behavior of PCa cellsThe levels of miR-203 in the tissues and paracancerous tissues of PCa and prostate cancer cell line DU-145 and human prostate epithelial cell line RWPE-1 were detected by qPCR,so called fluorescent quantitative PCR.The network-related database was used to predict the miR-203 downstream-related target gene RGS17,and the mutual binding relationship was confirmed by constructing a 3’UTR reporter gene plasmid containing RGS17,and interfering regulation in cells by luciferase reporter assay.The transfection efficiency of RGS17 was measured by qPCR,and the expression of RGS17 was measured by WB;the regulation of RGS17 by miR-203 was investigated.The effect of miR-203 on the value-added ability of prostate cancer cells was completed by MTT and clone formation assay;the cell migration and invasion assay was completed by transwell assay.part 3:Exploration of the effect of lncCAR10/miR-203 on the biological behavior of PCa cellsPaired prostate cancer and paraneoplastic normal tissue specimens were collected and the expression levels of Inc CAR 10 and miR-203 in prostate cancer and paraneoplastic tissues and prostate cancer cells and prostate cells were verified by real-time quantitative PCR(qPCR).To verify the effect of lncRNA CAR10 on the biological behavior of prostate cancer cells by inhibiting the target gene miR-203,dual luciferase reporter analysis was performed to detect the regulatory effect of IncRNA CAR 10 on miR-203 in prostate cancer cells;the effect on prostate cancer cell cycle and apoptosis was accomplished by flow cytometric assay;the detection of prostate cancer cell The MTT assay was applied to detect the proliferation of prostate cancer cells.Results:Regular consumption of green vegetables(>3 times per week)(x2=13.51,P=0.00)and regular physical activity(>3 times per week)(x2=45.55,P=0.00)benefited from prostate cancer prevention(P<0.05).Unhealthy lifestyle:regular smoking led to a significantly increased risk of prostate cancer(χ2=6.27,P=0.012).No statistically significant differences were found in the general clinical characteristics of the test subjects such as mean age,occupation,education level and BMI(P>0.05).The PSA level in the puncture biopsy-positive group was significantly higher than that in the puncture-negative group,which was statistically significant(P<0.05).This experiment confirmed the significant low expression of miR-203 in PCa tissues and PCa cell lines.Exogenous promotion of miR-203 expression in prostate cancer cells plays an oncogenic role was confirmed in in vitro experiments,while the present experiment showed that miR-203 has mutual target binding with the 3’UTR region of RGS17,which can inhibit the malignant progression of prostate cancer after RGS17 silencing,while the elevated level of RGS17 expression can contribute to Prostate cancer progression was promoted by elevated RGS17 expression.We found that lncCAR10 expression level was elevated in PCa tissues and miR-203 expression level was decreased.lncCAR10 expression level was significantly higher in PCa tissues and PCa cells than in normal prostate tissues and cells,and high expression of lncCAR1O could promote the malignant progression of PCa biological behavior.lncCAR1O effects on PCa cell biological behavior could be observed.The effect of lncCAR10 on the biological behavior of PCa cells could be achieved by downregulating the expression level of miR-203.Conclusion:Studies have shown that smoking is a risk factor for elevated prostate cancer;a lifestyle based on green vegetables and exercise is beneficial in reducing the risk of prostate cancer.The combination of PSA levels and dietary and lifestyle factors of the patients provides feasible support for tertiary prevention of PCa.It was demonstrated that inhibition of miR-203 expression promotes the expression of oncogene RGS17 can affect the biological behavior of prostate cancer.lncCAR10 has an effect on tumor cell-related biological behavior
