1.Cynanchum taihangense Tsiang et Zhang is an Asclepiadaceous plant and chiefly distributed in Shanxi Province,China.It grows in the bushes in the mountains.In our investigation,50 compounds were isolated from the 95%ethanol extract of the fresh plants of C.taihangense,38 of which were identified as cynataihoside A(1),cynataihoside B(2),cynanoside J(3),glaucoside H(4),cynataihoside C(5),cynataihoside D(6),cynanside A(7),cynatratoside C(8),glaucoside D(9),cynataihoside E(10),cynataihoside F(11),glaucoside I(12),stauntoside L(13),cynataihoside G(14),cynataihoside H(15),cynataihoside I(16),cynataihoside J(17),cynataihoside K(18),cynataihoside L(19),cynatratoside E(20),glaucoside A(21),atratcynoside F(22),cynataihoside M(23),cynataihoside N(24),cynataihoside O(25),cynataihoside P(26),cynataihoside Q(27),cynataihoside R(28),cynataihoside S(29),cynataihoside T(30),α-viniferin(31),syringaresinol(32),syringaresinol O-β-D-glucopyranosid(33),1-(4-hydroxy-3-methoxyphenyl)-1,2-propanediol(34),2,5-dihydroxyacetophenone(35),pyroglutamate(36),4-hydroxyacetophenone(37),vanillic acid(38).Among them,1-2,5-6,10-11,14-19,23-29 and 30 were new compounds,and the aglucone of 1,11,14,26-27,29 and 30,totally five kinds,have not been reported in literature.The other compounds were isolated from C.taihangense for the first time.Thirteen compounds were subjected to detect the cytotoxicity against three human tumor cell lines(HL-60,THP-1 and Caco-2)using MTT method.The tested compounds showed a different degree of inhibitory activity against HL-60,THP-1 and Caco-2 cells.Compounds 2 and 8 displayed significant and selective cytotoxic activity on Caco-2 cell,respectively,and 2 was more effective than the positive control.Compound 5 was more sensitive to THP-1 cell line than HL-60 and Caco-2 cell lines with IC50 values of 7.85 μM.Moreover,the structure-activity relationship of the C21 steroidal glycosides on the cell lines was analyzed.2.The metabolism of secalonic acid D in SD rats was studied.The metabolites were collected and isolated from the urine of SD rats after administering secalonic acid D.Five compounds were obtained from the urine,and one of them was identified as 1H-indol-3-ol,3-(hydrogen sulfate)(SUM1).The obtention of 1H-Indol-3-ol,3-(hydrogen sulfate)(SUM1)may be attribute to the abnormal metabolism caused by secalonic acid D. |
