Objective: Long non coding RNA(lncrna)plays an important role in regulating the occurrence and development of glioma.Linc01116 is highly expressed in a variety of tumors and promotes the occurrence and development of tumors.However,its function and regulatory mechanism in gliomas are still unclear.In this study,we detected the expression and biological function of linc01116 in gliomas,to explore the mechanism of linc01116 in gliomas,to clarify the clinical significance of high expression of linc01116 in gliomas,and to evaluate the possibility of linc01116 as a potential drug target for gliomas.Methods: 1.The correlation between the expression of linc01116 in different grades of glioma and clinical prognosis was analyzed by bioinformatics technology,and the expression level of linc01116 in different grades of glioma tissues or glioma cell lines was detected by quantitative real-time polymerase chain reaction(q RT-PCR),and the correlation between the expression of linc01116 and glioma proliferation was analyzed.2.Lentiviral vector was used to construct the glioblastoma cell lines with stable overexpression(OE)or knockout(KD)of linc01116.CCK8,colony formation and Transwell migration assay were used to detect the effects of linc01116 OE or KD on the viability,proliferation and migration of glioblastoma cells.3.The correlation between linc01116 and MDM2-p53 pathway key genes was analyzed by bioinformatics technology,and the effects of linc01116 oe or KD on the m RNA and protein expression of MDM2-p53 pathway key genes in glioma cell lines were detected by q RT-PCR and WB experiments,so as to clarify the correlation between linc01116 and MDM2-p53 pathway in glioma and whether linc01116 indirectly affects p53 and its target by regulating the expression of MDM2 Gene expression.4.The potential mechanism of linc01116 regulating MDM2-p53 signaling pathway was predicted by bioinformatics technology,and the correlation between linc01116 and mirna-744-5p and MDM2 was verified by RT q PCR,WB,luciferase reporter gene and rip,respectively,so as to determine whether linc01116 can competitively bind mir-744-5p with MDM2 m RNA through Cerna mechanism to affect the expression of MDM2 in glioma cells.5.The expression and function of mir-744-5p in gliomas were detected by q RT-PCR and CCK8,and the correlation between mir-744-5p and gliomas proliferation ability and linc01116 expression was analyzed;whether mir-744-5p can reverse linc01116 proliferation and migration ability was detected by rescue experiment.6.The effect of linc01116 kd on the tumorigenicity of glioma was detected by nude mice tumorigenesis experiment;the effect of linc01116 kd on the proliferation of tumor cells was detected by TUNEL analysis;whether linc01116 kd can regulate mirna-744-5p-mdm2-p53 was further confirmed by q RT-PCR and WB experiment.Results:1.Overexpression of linc01116 in gliomas was negatively correlated with malignant phenotype and survival time of patients;2.Overexpression of linc01116 in gliomas promoted cell viability,colony formation and migration;conversely,knockdown of linc01116 inhibited cell viability,colony formation and migration;3.Linc01116 and MDM2-p53 signaling pathway in gliomas Linc01116 can regulate MDM2-p53 signaling pathway and its target genes by inhibiting the expression of MDM2 m RNA;4.Linc01116 can competitively bind mirna-744-5p with MDM2 m RNA through Cerna mechanism in gliomas to block the silencing effect of mirna-744-5p on MDM2 m RNA;5.Mirna-744-5p is a tumor suppressor in gliomas,which is low expressed in gliomas and closely related to linc0116.In vivo experiments confirmed that linc01116 kd could inhibit the tumorigenicity and proliferation of glioma cells,and further confirmed that linc01116 could regulate the function of mirna-744-5p-mdm2-p53 in glioma.Conclusion: linc01116 is highly expressed in gliomas,which is related to the malignant phenotype of gliomas,and is an important tumor promoting factor;linc01116 can regulate mirna-744-5p-mdm2-p53 through the mechanism of Cerna in gliomas,thus promoting the proliferation and migration of gliomas;mir-744-5p is low expressed in gliomas,which has anti-cancer effect and can inhibit the tumor promoting effect of linc01116;linc01116 can inhibit the tumor promoting effect of linc01116 In conclusion,linc01116 can be used as a potential target for glioma therapy.
