| Background$Aims:Primary liver cancer is the sixth most common cancer and the third most common cause of death in cancer.70-90%of primary liver cancer is hepatocellular carcinoma(HCC).Liver resection is still being considered as first-line treatment of HCC.There are different opinions about the benefits of transarterial chemoembolization(TACE)for patients receving hepatectomy.Although TACE can benefit some patients,but there is still a lack of effective biological markers to predict response to treatment of TACE.There would be side effects of elevated alanine aminotransferase(ALT)and myelotoxicity after TACE treatment,and it is still a lack of effective biomarker to predict the side effects of TACE.Methods:We recruited 485 HCC patients from The First Affiliated Hospital,Guangxi Medical University.Tissue samples from HCC lesions were obtained after surgical resection,immediately snap-frozen,and stored at-80°C until needed for DNA extraction.Polymorphisms showing genotyping with call rates less than 95%,violation of Hardy-Weinberg equilibrium(HWE)(P<1.0×10-6),or minor allele frequencies less than 5%in both cases and controls were excluded from the analysis.Exome data were analyzed by R,Plink and EPacts3.2.6.Fine mapping of genes were performed by Locus Zoom Packages.For the candidate genes to be verified,lentivirus was used to interference the expression of selected genes.For SNPs to be verified,the genotypes of selected SNP were genotyped by Sanger’s methods,chemotherapy drugs were used to interference hepatic cell lines.The ALT levels were valued by enzyme linked immunosorbent assay(Elisa).The univariate analysis between clinical features and overall survival was com Pared using the log-rank test,and those with P<0.1 were entered into the Cox proportional hazard regression model.A P-value<0.05 was defined as statistically significant.All statistical analyses were performed using SPSS 16.0(SPSS,Inc.,Chicago,IL,USA).Results:Overall survival of 485 patients was associated with postoperative adjuvant TACE(log-rank P=0.043).Expression of ILF3 was associated with cell apoptosis,invasiveness and Proliferation(P<0.05).Genotypes of rs2239560were associated with elevated alanine aminotransferase(ALT)and myelotoxicity after TACE treatment(P<0.05).Coclusion:Postoperative adjuvant TACE does not improve long-term prognosis of patients,but improve the short-term prognosis.Expression of ILF3 gene can affect the prognosis of postoperative adjuvant TACE.SNP rs2239560 was associated with elevated alanine aminotransferase(ALT)and myelotoxicity after TACE treatment. |
