PurposeEpigenetic alteration may play an important role in development of thyroid cancer.Involvement of long noncoding RNAs in regulation of cell proliferation,migration and invasion in thyroid papillary carcinoma(PTC)has been reported.The role of Braf-activated non-coding RNA(BANCR)in PTC is controversial.The purpose of this study is to explore the role and molecular mechanism of BANCR in PTC.MethodsThe expression of BANCR was detected by q PCR in thyroid nodules and cell lines.Cell proliferation,migration and invasion experiments and the subcutaneous implant tumor model of nude mice were conducted to evaluate the effect of BANCR on the proliferation,migration and invasion of PTC.Target molecule of BANCR was confirmed by RNA pulldown and RNA immunocoprecipitation assay.The effects of ANXA2 on PTC were assessed by tumor proliferation,invasion and migration assays and subcutaneous implant tumor model in nude mice in vivo.The protein levels and phosphorylation levels of protein related to MAPK/ERK pathway,Wnt/β-catenin pathway,as well as cell proliferation marker and epithelial mesenchymal transformation markers,were detected by Western blot.ResultsBANCR is highly expressed in PTC tissues and cells,and higher in those with BrafV600E mutation.BANCR combines with ANXA2 protein and promotes the proliferation of PTC dependent on mutated Braf protein.MAPK/ERK pathway was involved in this process.At the same time,BANCR and ANXA2 promote the migration and invasion of PTC and the process of epithelial mesenchymal transformation.Wnt/β-Catenin pathway is involved in this process.ConclusionLncRNA BANCR,as a cancer-promoting gene,promotes the progress of PTC together with ANXA2 protein.This study provides a new research direction for the diagnosis,treatment and monitoring of PTC.
