Objective:1.To observe the characteristics of HLP model prepared by high cholesterol and high fat diet feeding method,and the regulating effect of medicine cake moxibustion on blood lipid of HLP model rabbits.2.To observe whether the influence of medicine cake moxibustion on HLP model rabbits is involved in Wnt-SREBP /HMGR related pathway.3.To observe the influence of medicine cake moxibustion on lipid metabolism of HLP model rabbits.Methods:1.After adaptive feeding for one week,72 male New Zealand rabbits were randomly divided into six groups,normal group,model group,medicine cake moxibustion group,western medicine group,inhibitor group and normal saline group.After feeding high-cholesterol and high-fat formula for 8 weeks,the groups were fixed and bundled according to the factors of the subjects,respectively,moxibustion with drug cake,feeding Atorvastatin calcium,medicine cake moxibustion and intraperitoneal injection of doxorubicin or injection of normal saline.The intervention lasted for 8 weeks.The blood lipid levels in each group were detected before modeling,4 weeks after modeling,8 weeks after modeling,and 8 weeks after intervention.2.After 8 weeks of intervention,serum,liver tissue and myocardial tissue samples were taken from each group,and related pathway indexes were detected by ELISA,WB,and RT-PCR techniques.3.After 8 weeks of intervention,rabbit plasma of the normal group,the model group and the medicine cake moxibustion group were taken to analyze the differences of lipid metabolites between the groups by LC/MS method.Results:1.Before modeling,there was no statistical significance in blood lipid levels among all groups(P>0.05).After 8 weeks of modeling,compared with the normal group,serum TC content in model group,medicine cake moxibustion group and western medicine group increased(P<0.01,P<0.01,P<0.001),LDL content increased(P<0.01,P<0.001,P<0.05),and TG content increased(P<0.001,P<0.01,P<0.01).There were no significant differences in serum TC,LDL and TG contents among the model groups(P>0.05).After 8 weeks of intervention,compared with the model group,the contents of serum TC,LDL and TG in the medicine cake moxibustion group decreased(P<0.001,P<0.01,P<0.001),and the contents of serum TC,LDL and TG in the western medicine group decreased(P<0.001,P<0.05,P<0.001).There was no statistical significance in serum TC,LDL and TG contents between the medicine cake moxibustion group and the western medicine group(P>0.05).2.Compared with normal group,the contents of IGF and SP1 in serum of model group and inhibitor group were increased(P<0.01).Compared with model group,the contents of IGF and SP1 in serum of medicine cake moxibustion group,western medicine group and normal saline group were decreased(P<0.01).Compared with the medicine cake moxibustion group,the serum IGF content in the inhibitor group was increased(P<0.01).Compared with model group,the relative expression of HMGR in liver tissue of medicine cake moxibustion group,western medicine group and normal saline group decreased(P<0.05).Compared with normal group,the mRNA expression of SREBP-1c in liver tissue of model group was increased(P<0.001).Compared with the model group,the expression level of the medicine cake moxibustion group and the normal saline group decreased(P<0.001),and the expression level of the western medicine group decreased(P<0.01);Compared with normal saline group,the expression level of inhibitor group was increased(P<0.05).3.There was a great difference in lipid metabolism between the normal group,the model group and the moxibustion group with medicine cake isolation.The top 15 metabolites were all phospholipids,mainly PC,SM and LPC,and the concentration of metabolites from high to low were: the model group > medicine cake moxibustion group >normal group.Conclusion:1.The preparation of HLP model with high cholesterol and high fat diet can form the model in 4 weeks and stabilize relatively in 8 weeks.The medicine separation cake moxibustion can reduce the level of blood lipid in HLP model rabbits,mainly the level of serum TG.2.Herb medicinal cake moxibustion down-regulates the contents of SREBP-1C and HMGR in liver tissue,and may regulate the synthesis of fat(TG)and cholesterol(TC)in HLP model rabbits through this way.3.The down-regulation of SREBP-1C in liver tissue by herb medicinal cake moxibustion may be caused by down-regulation of PC phospholipid metabolites. |
