| Objective:Understand DNER protein expression in a variety of tumor tissue,and by observing the effect on prostate cancer cell proliferation and migration ability to provide theoretical reference to the research of breast cancer,further explore DNER protein expression in breast cancer and prognosis,through cytology test observation DNER influence on breast cancer cell proliferation and migration ability,and study its specific mechanism,DNER may become breast cancer new therapeutic targets for the future to improve theoretical basis.Method:Methods:Using reverse transcription polymerase chain reaction(RT-PCR)to detect different tumor tissues of DNER mRNA expression,using immunohistochemical method to detect the expression of prostate cancer and normal tissue and analyze its relationship with the clinical pathological data,to cut the DNER PC3 prostate cancer cells were determined by MTT experiment,scratches,Transwell experiment,cell cycle detection to clear DNER impact on prostate cancer cell proliferation and migration ability.Using immunohistochemical and quantum dot markers detection DNER protein expression and analysis of clinical data,rt-pcr and Western blot detection of breast cancer cell line DNER and DNER mRNA expression level of cells,screening for breast cancer cells MDA-MB-231 DNER downgrade were determined by MTT experiment,scratches,Transwell experiment,cell cycle detection to clear DNER influence on breast cancer cell proliferation,migration ability,IPA method analysis of signaling pathways,RT-PCR and Western blot test access key protein expression.Results:Malignant tumor tissues to detect DNER mRNA expression level,compared with the normal tissue in non-small cell lung cancer,and pancreatic cancer was found in DNER mRNA expression level increased,but did not see the differences in the brain tumor.DNER protein in prostate cancer tissue increased,and the distant metastasis of prostate cancer were positively correlated,the probability of distant metastasis positive expression is in a larger,especially related to bone metastases,further to downgrade DNER PC3 prostate cancer cells were determined by MTT experiment,found that relative to the ability to control its growth has fallen;The cell cycle was changed,and the proportion of G1 phase increased significantly,while the proportion of S phase and G2/M decreased.The change of the migration ability was detected by the scratch test,and the migration ability was decreased compared with the control group,and the same result was found in the Transwell experiment.The results showed that the gene expression and protein expression of HES1 and GLI1 were decreased,but it had no effect on HEY1.DNER organization high expression in breast cancer,is associated with lymph node,HER-2,PR,further to downgrade DNER of breast cancer cells were determined by MTT experiment results growth ability drops,scratches,Transwell experiments and experimental results for migration ability,IPA method test results of PI3K/AKT signaling pathways such as expression,western blot and RT-PCR detection DNER cut after the results of the MDA-MB-231 cells for Girdin protein and mRNA level have no obvious changes,phosphorylated AKT significantly reduced,and P85a also significantly reduced.Conclusion:DNER protein in a variety of malignant tumor tissue increased,and is one of the independent prognostic indicators for prostate cancer,it can promote the ability of prostate cancer cell proliferation and migration,and inhibiting apoptosis.The high expression of DNER protein in breast cancer tissues is positively correlated with the occurrence of breast cancer,and the high expression of DNER in breast cancer patients has a lower survival rate and is one of the independent prognostic indicators of breast cancer.DNER similarly high expression in breast cancer cell lines,MDA-MB-231 cells expressed in the highest,DNER can promote breast cancer cells value-added,migration ability,mainly through the PI3K/AKT signaling pathways regulate breast cancer cells of stem cell properties,while Girdin protein may be the key factor of the regulation.DNER could be a new target for breast cancer treatment. |
