| Objective:To observe the effects of Modified decoction of rehmanniae on D-gal induced subacute aging related gene mRNA and protein memory ability,antioxidant ability and hippocampus expression in mice,to investigate the effect and mechanism of Modified decoction of rehmanniae Treating aging.To observe the effect of Modified decoction of rehmanniae on learning and memory ability,antioxidant ability,related gene and protein expression in hippocampus of D-gal-induced aging mice,and to explore the effect and mechanism of Modified decoction of rehmanniae on senescenceMethods:1.105 male ICR mice(1 month old mice,15 mice,7 month old mice 90)were divided into youth group,middle age group,middle-aged VE group,middle-aged Rehmannia group,senescence group,aging VE group,and senile Rehmannia glutinosa group,7 groups.D-gal was injected intraperi-toneally at the dose of 1000mg kg/d for six weeks everyday.Normal feeding of mice during the molding process.The model was built,1 days after intragastric administration of Modified decoction of rehmanniae and VE.According to VE 250mg kg/d dose gavage of Modified decoction of rehmanniae according to 20.6g kg/d dose administered,youth group,middle-aged and aged groups by intragastric gavage corresponding volume of saline,around,after the Morris water maze test,finally drawn.2.The level of mRNA expression of Wnt-3a,GSK-3 β,C-myc,TCF-4 and β-catenin in hippocampus was detected by Real-time PCR.3.Western blot was used to detect the expression level of Wnt-3a,GSK-3 β,C-myc,TCF-4 and β-catenin in the hippocampus.Results:1.Morris water maze navigation experiment results showed that the mice for several days of swimming training,Modified decoction of rehmanniae can make mice escape latency gradually shortened;in the space exploration experiment,Modified decoction of rehmanniae can significantly increase the number of D-gal induced subacute aging mice through the security platform area,compared with the aging group had significant difference(P<0.01).2.The expression of mRNA gene in hippocampus:detected by Real-time PCR method showed that Modified decoction of rehmanniae can up regulate the expression of D-gal in hippocampus in subacute aging mice induced by Wnt-3a,TCF-4,β-catenin gene mRNA in mouse aging process,inhibit the expression of GSK-3 β,C-myc gene mRNA.3.Gene expression in the hippocampus:detected by Western blot method showed that Modified decoction of rehmanniae can up regulate the expression of D-gal in hippocampus in subacute aging mice induced by Wnt-3a,TCF-4 and β-catenin gene protein in mouse aging process,inhibit the expression of GSK-3β and C-myc protein.Conclusions:1.Modified decoction of rehmanniae can improve the cognitive ability of D-galactose-induced aging mice.2.Modified decoction of rehmanniae can promote the differentiation of neural stem cells in the brain,increase the expression of wnt-3a,TCF-4 andβ-series genes and their proteins,and reduce the expression of GSK-3β,C-myc gene and its protein.3.Modified decoction of rehmanniae can play a therapeutic role in aging mice induced by D-gal by activating the WNT signaling pathway. |
