Objective:The seeds of Descurainia Sophia(D.Sophia)belongs to the genus Cruciferous,of which were important material in tradition-al Chinese medicine,the plant is widely distributed more than 10 provinces in China.Modern pharmacological studies had shown that it had strong activities,such as anti-ulcer,anti-aging,anti-diabetic,anti-tussive,expectorant,anti-asthmatic,throat disease,measles and smallpox activity.Study of the chemical constituents of the seeds of D.Sophia can furtherclarify the basis of its efficacy substances and find new active ingredient resources,which lay the foundation for its further development and utilization.Methods:1.The seeds of D.Sophia were extracted by refluxing with water three times and filtered.The combined extracts were concentr-ated under reduced pressure in a vacuum evaporator to the gross extracts.They were dissolved in water,yield the water soluble matter and the water insoluble matter,and then the water soluble matter were subjected to Diaion HP-20 porous polymer resin and eluted with H2O,10%CH3OH,30%CH3OH,50%CH3OH,1 00%CH3OH,successi-vely.The fractions eluted were subjected to Toyopearl HW-40 column chromatography and eluted successively to afford subfractions.Then the subfractions were isolated and purified by Toyopearl HW-40,Sephadex LH-20,silica gel column chromategraphy,preparative liquid chromatography and recrystallized to give compounds.The structures were identified on the basis of physiochemical characteris-tics and spectral methods such as IR,UV,1H-NMR,13C-NMR,1H-1H COSY,DEPT-135,HMBC,HSQC,and so on.2.(1)The main goal of this study was to obtain the maximum yield of the oil by an optimal selection of supercritical fluid extraction parameters.According to the central composite design and response surface methodology for supercritical fluid extraction method,a quadratic polynomial model was used to predict the yield of D.soph-ia seed oil.A series of runs was performed to assess the optimal ext-raction conditions;(2)The analysis of fatty oil in the seeds of Lepidi-um apetalum(L.Lepidium)and D.Sophia provides an experimental basis for further research on the fatty oil component for the first time by using the supercritical CO2 extraction method;(3)Two different habitats of the seeds of D.Sophia were extracted by supercritical CO2 extraction method and analysed by GC-MS and showed differences and compositions.3.A resolution liquid chromatography coupled with electrospray ionization quadrupole time-of-flight tanden mass spectrometry(ESI-TOF MS/MS)spectrum,fragmentation reactions of the[M-H]-。Consults:1.35 compounds were isolated from the seeds of D.Sophia and elucidated by spectrosopic and chemical methods.Seven new compounds:They were Deseurainiaside A(TLZ-11-1-1),Deseurainia-side B(TLZ-11-1-2),Deseurainiaside C(TLZ-11-1-4),Deseurainia-side D(TLZ-11-1-5),Deseurainiaside E(TLZ-11-1-7),Deseurainia-side F(TLZ-12-10),Deseurainiaside G(TLZ-19-6);The other ten known compoundsisolated from this plant for the first time:4’-metho xy-kaempferol-3-O-β-D-glucopyranosy-7-O-β-gentiobioside(TLZ-5-1-1),kaempferol-3,7-di-O-β-D-glucoyranoside(TLZ-23-10),salicyl-icacid(TLZ-16),cyclo-(L-Trp-D-Val)(TLZ-19-5),artabotryside-A(TL Z-17-2),isofraxidin(TLZ-14-8),lyoniresinol(TLZ-5-C),octadecane(TLZ-18),1’-O-benzyl-α-rhamnopyranosyl-(1→6)-β-D-glucopyra-noside(TLZ-27),3,5-dihydroxyphenethyl alcohol-3-O-β-D-glucopy-ranoside(TLZ-29).2.(1)The results indicated that the extraction pressure had the greatest impact on oil yield within the range of the operating conditions studied;(2)GC-MS was used to analyse the chemical composition of the seeds of D.Sophiaand L.apetalum and which was found to have a very complex composition with compound identifica-tions reported;(3)Two different habitats of the seeds of D.Sophia were analysed by GC-MS and show differences and compositions.Although the number of components are different,but the main prod-uct is the same,but there is large difference in the percent content.3.3 7 compounds including 10 target compounds and 27 nontarget compounds were screened out in 70%ethanol extract of the seeds of D.Sophia which 18 of them were found to be possible novel compounds and 7 of them found for the first time.
