Objective:Through behavioral observation,transmission electron microscope observation and the expression of GABAa,BDNF,Trk B,SYN,GAT-1,PSD95 protein and m RNA in the cerebral cortex and hippocampus,the interventional effects of electroacupuncture Quchi and Yanglingquan on SCA rats were studied.Reveal its mechanism of regulating synaptic plasticity to relieve the spasticity of stroke limbs.Methods:77 male SD rats were randomly divided into blank control group and sham operation group,and the remaining 59 rats were entered into the model reserve group.Zea-longa thread embolization method and intracapsular injection of NMDA receptor were used to prepare a rat model of stroke limb spasm.After death and behavioral testing,18 rats with successful modeling were randomly divided into model group and electroacupuncture group(bilateral Yanglingquan and Quchi line electroacupuncture,30 min/time,once/d for 5 days).Behavioral observation before and after treatment(Zea-longa thread plug score for neurological function score,modified Ashworth muscle tone scale for muscle tone assessment);using electron microscope to observe the microscopic condition of synaptic neurons in the affected cerebral cortex and hippocampus;Western Blot was used to detect the expression of GABAa,BDNF,Trk B,SYN,PSD95,and GAT-1 protein in the affected cerebral cortex and hippocampus by immunoblotting.Real-time quantitative PCR was used to detect GABAa,BDNF,Trk B,and SYN in the affected cerebral cortex and hippocampus.,PSD95,GAT-1 m RNA expression.Results:1.Behavioral testing:(1)Zea-longa nerve function score: Compared with the blank control group and sham operation group,the model group score increased(P<0.01);compared with the model group,the electroacupuncture group scoredecreased(P<0.05).(2)Modified Ashworth muscle tone score: Compared with the blank control group and sham operation group,the model group score increased(P<0.01);compared with the model group,the electroacupuncture group score decreased(P<0.05).2.Observation of ultrastructure changes of synapses in rat brain by electron microscope: Compared with blank control group and sham operation group,the number of synapses in the model group decreased significantly,and the density of vesicles decreased unevenly in the cortex and hippocampus.The fusion boundary between the membrane and the gap is blurred,the post-synaptic cell terminal cytoplasmic degeneration and dissolution are significant,and the density of the post-synaptic dense band also decreases;the number of synapses in the electroacupuncture group is significantly increased and relatively dense,and the number of vesicles is increased.The newly-increased concave synapse increases,the dense body thickens after the synapse,and the gap becomes regular and tight.The intra-cortical recovery of the electroacupuncture group was better than that of the hippocampus.3.Western Blot detection by immunoblotting: Compared with the blank control group and the sham operation group,the expression of GABAa,BDNF,Trk B,SYN,and PSD95 receptor protein in the model group was reduced(P<0.01,P<0.05).The expression of GAT-1 receptor protein was increased(P<0.01,P<0.05);compared with the model group,the expression of GABAa,BDNF,Trk B,SYN,PSD95 receptor protein was increased in the electroacupuncture group(P<0.01,P< 0.05),while GAT-1 receptor protein expression decreased(P<0.01,P<0.05).Related expression results in hippocampus are consistent with cortex.4.Fluorescence real-time quantitative PCR detection: In the cortex,compared with the blank control group and sham operation group,the model group GABAa,BDNF,Trk B,SYN,PSD95 m RNA expression were reduced(P<0.01,P<0.05),while GAT-1 m RNA expression increased(P<0.01,P<0.05);compared with the model group,GABAa,BDNF,Trk B,SYN,PSD95 m RNA expression increased in the electroacupuncture group(P<0.01,P<0.05),while GAT-1 m RNA expression decreased(P<0.01,P<0.05).Related expression results in hippocampus are consistent with cortex.Conclusion:1.Zea-longa thread embolization method + intracapsular injection of NMDA receptor to construct a mold for stroke limb spasm has certain feasibility.2.Electro-acupuncture Quchi and Yanglingquan significantly improved the nerve injury and limb spasm in SCA rats.3.The mechanism of intervention effect of electroacupuncture Quchi and Yanglingquan may be achieved by regulating the synaptic plasticity of the brain.
