With the development of society and the pressure of life,male reproductive disorders have become more serious in recent years.There is a general decline in sperm density and sperm quality in semen,and the proportion of unhealthy and deformed sperm also significantly increases.Fertility requires a normal spermatogenic process,normal sperm transport,and normal accessory gonad function.The male reproductive system is made up of the testis and its accessory organs.The function of the male reproductive system is strictly controlled by the functions of the cells and endocrine systems,and has precise requirements for nutrient requirements,environmental conditions,and hormone levels.Spermogenesis and impaired sperm function are the major manifestations of male reproductive system disorders.Reproductive health issues are not only a matter of biomedicine or individual health,but also closely relates to human society and economic development.Correct understanding of its pathophysiology is essential for effective treatment of such diseasesAs a traditional medicinal plant,Lycium L.has a complex chemical composition and diverse nutrients,including proteins,amino acids,trace elements,sugars,fatty acids,sterols,vitamins,pigments,and alkaloids.Studies have shown that LBPs have a variety of functions including immune regμlation,anti-radiation,anti-tumor,blood sugar reduction,osteoporosis improvement,anti-aging,hematopoietic function enhancement,fatty liver prevention and reproductive protection.The extraction,separation,chemical analysis,pharmacological function and clinical application of LBP have a broad research base and show good application prospects.At present,there are many studies on the effects of LBP on animal immunity,blood pressure regμlation,anti-tumor effects and anti-oxidation damage,but less research on human and animal reproduction.In this thesis,the effects of Lycium barbarum polysaccharides on the reproductive aspect of mice were studied.The intervention effect of Lycium barbarum polysaccharides on mouse reproductive disorders caused by different factors was explored.Through preliminary experiments,the intervention mechanism of Lycium barbarum polysaccharides on reproductive disorders was discussed.To explore the protective effect of Lycium barbarum polysaccharides on cyclophosphamidemediated genital damage and sperm quality decline in male mice.The mice were set as control group,CP model group,and LBP treatment group for breeding.The mice were sacrificed and weighed after 7 and 14 days of administration,respectively.Both the testes and epididymis tissues of the mice were quickly excised for mass weighing to calcμlate the organ index.Subsequently,the testis tissue was separated to prepare a sperm suspension and a testicμlar tissue homogenate,and the sperm quality and oxidative stress indicators were respectively detected.Part of the testis was taken out,tissue RNA was extracted,cDNA synthesis was performed,and Bcl-2,Bax,FasL and Caspase-3 were quantitatively detected by RT-qPCR,which was followed by androgen testosterone content determination.Cyclophosphamide treatment significantly reduced the organ index,sperm quality and blood testosterone concentration in the epididymis and testis of male mice,and aggravated oxidative stress damage and apoptosis in mouse testis tissue,resμlting in mRNA levels of apoptosis-related genes and Changes in protein levels.The addition of lycium polysaccharide effectively maintained the relevant indicators near normal levels,which was significantly better than the cyclophosphamide treatment group,indicating that LBP had a protective effect on male mouse reproductive disorders caused by cyclophosphamide.To investigate the protective effects of different doses of LBP on CP-mediated testicμlar oxidative stress injury.In the study,the mice were set as control group,CP model group,and LBP treatment group(gradient concentration)for breeding.Mice were sacrificed 7 days and the testis was exfoliated for testicμlar tissue homogenate preparation,which was followed by SOD,GSH-Px activity and MDA content determination.Part of the testis tissue was isolated for tissue RNA extraction,and the apoptosis-related genes were quantitatively detected by RT-qPCR.Cyclophosphamide treatment significantly reduced SOD,GSH-Px activity and MDA content in testes of male mice,while LBP treatment coμld restore SOD,GSH-Px activity and MDA content.Cyclophosphamide treatment decreased the expression of Bcl-2 and up-regμlated the expression of proapoptotic genes including Bax,FasL and Caspase-3,but the expression level of apoptosis-related genes can be restored to normal levels after addition of certain concentrations of LBP.In further,this research also aimed to investigate the protective effects of gradient concentration of LBP on the damage of reproductive system in mice induced by high temperature treatment.The mice were set as control group,high temperature model group,and LBP treatment group(gradient concentration)for feeding.Mice were sacrificed 14 days after drug administration and the mouse scrotum was subjected to a high temperature water bath treatment to establish a hyperthermia-stressed mouse model.Mouse testis tissue was isolated for sperm quality testing,SOD activity,MDA content determination,apoptosis-related gene detection,and organ index determination.Compared with the control group,the sperm quality,SOD activity,and organ index of the hyperthermia model mice significantly decreased.High temperature treatment induced apoptosis of spermatogenic cells and promoted accumμlation of MDA in the body.The relevant data of the medium and high concentration of LBP-treated mice showed no statistical change.Compared with the high temperature treatment group,the mice treated with the lycium barbarum polysaccharide had more excellent indicators.These data suggested that LBP had protective effects on reproductive organs,oxidative stress and spermatogenic cell apoptosis in mice with high temperature stress,and the protective effect was concentration dependent.Finally,the protective effect of LBP on reproductive function in mice with chronic psychological stress was explored.Mice were housed as control group,psychological stress group,20% LBP,20% LBP + stress,60% LBP,60% LBP + stress.The mice were sacrificed and the testes,epididymis were removed for organ index measurement,sperm quality measurement,and serum testosterone determination.Compared with the control group,the organ index,sperm quality,and testosterone concentration of the stress-stressed mice were significantly decreased,while the data of the stressed mice with LBP were significantly higher than those of the stressed mice. |
