Metabolomics Prolifing Of Rhegmatogenous Retinal Detachment Using Vitreous Fluid | | Posted on:2017-06-02 | Degree:Doctor | Type:Dissertation | | Country:China | Candidate:X M Lu | Full Text:PDF | | GTID:1364330590491845 | Subject:Ophthalmology | | Abstract/Summary: | PDF Full Text Request | | Purpose: To evaluate whether idiopathic macular hole is appropriate control for metabolomics study,and to analysis the metabolic characteristic of rhegmatogenous retinal detachment using vitreous fluid.Methods: 1.1H-Nuclear Magnetic Resonance specra were acquired from vitreous samples of eight idiopathic macular hole patients and five cornea donor eyes.SIMKA-P+12.0 was applied for data analysis.Principal component analysis,partial least square-discriminant analysis and orthogonal partial least squares discriminant analysis were used for pattern recognition.Variable importance in the projection based on orthogonal partial least squares discriminant analysis model was used to rank the importance of metabolites.2.Twenty vitreous samples of rhegmatogenous retinal detachment patients were obtained for metabolomics study.Patients were divided into two groups according to proliferative vitreoretinopathy classification.Sixteen proliferative vitreoretinopathy grade B patients were compared with cornea donor eyes or idiopathic macular holes of same gender.Four proliferative vitreoretinopathy grade C patients were compared with both cornea donor eyes and idiopathic macular holes.Metabolic profiles were acquired and analyzed as above.Results: 1.The metabolic models of idiopathic macular hole patients and cornea donor eyes were statistically significant.Twenty-one metabolites were detected in the vitreous sample and nine of which showed statistically significant changes.Main changes were sugar metabolism and amino acid metabolism.2.No statistically difference between the metabolic models of proliferative vitreoretinopathy grade B rhegmatogenous retinal detachment patients and idiopathic macular hole patients.The metabolic models of proliferative vitreoretinopathy grade B rhegmatogenous retinal detachment patients and cornea donor eyes were statistically significant.The metabolic models of proliferative vitreoretinopathy grade C rhegmatogenous retinal detachment patients were statistically significant with both idiopathic macular hole patients and cornea donors.Conclusion: 1.Statistically significant metabolic changes between idiopathic macular hole and cornea donor eyes discovered by 1H-nuclear magnetic resonance.Idiopathic macular hole is not suitable control for metabolomics study.2.Proliferative vitreoretinopathy grade B and C rhegmatogenous retinal detachment patients had similar metabolic trends.Metabolites related to energy metabolism,inflammation and glial cell proliferation were significantly altered. | | Keywords/Search Tags: | rhegmatogenous retinal detachment, proliferative vitreoretinopathy, vitreous, metabolism, nuclear magnetic resonance spectroscopy | PDF Full Text Request | Related items |
