ObjectivePsoriasis is a chronic skin disease characterized by hyperproliferation and abnormal differentiation of keratinocytes.The existing medical treatments have not achieved satisfactory results in the treatment of psoriasis.The natural small molecule compounds isolated from Chinese herbal medicines have a mount of biological activation with little side effect.Professor Lu Chuanjian from Guangdong Provincial Hospital of Traditional Chinese Medicine learn from Master of Chinese Medicine Guowei Xuan and optimize the in-hospital preparation of Yinxieling tablets of Guangdong Provincial Hospital of Traditional Chinese Medicine for the treatment of psoriasis into PSORI-CMO2.Gallic acid is a natural small molecule compound isolated from the PSORI-CMO2.This article studies the role of gallic acid in the treatment of psoriasis and describes its mechanism.MethodsIn vitro studies of gallic acid in the treatment of psoriasis.IL-17A induces human keratinocytes to establish a psoriasis-like cell model,and gallic acid treats the cell model.The effect of gallic acid on keratin 6,16,17 and signaling pathway Nrf2 was examined.In addition,the Nrf2 plasmid was transfected into human keratinocytes,and gallic acid treats the psoriasis-like cell model with high expression of Nrf2,and then the effect of gallic acid on keratin 6,16 and 17 was observed.In vivo studies of gallic acid in the treatment of psoriasis.Imiquimod induced BALB/c mice to establish psoriasis-like mouse model,and gallic acid treats the mice model.Psoriasis-like characterization,epidermal thickening,expression of keratin 6,16,17 and signaling pathway Nrf2 were measured.ResultsGallic acid significantly inhibits the expression of keratin 6,16,17 and the activiation of signaling pathway Nrf2 in the psoriasis-like cell model and in the psoriatic-like mouse model.In vitro,sustained high expression of Nrf2 counteracted the effect of gallic acid on the inhibition of keratin 6,16 and 17 expression.ConclusionGallic acid inhibits the expression of keratin 6,16,17 through Nrf2 in the treatment of psoriasis-like disease.
