Nonarteritic anterior ischemic optic neuropathy(NAION)is a common clinical optic nerve disease,old ages are more susceptible.The artery ischemia of the anterior optic nerve blood supply lead to insufficient blood perfusion pressure and disease,and make serious damage to visual function.There is no generally accepted treatment for the unknown Pathogenesis.Tutor combined the theory of qi and blood ophthalmology syndrome differentiation with the theory of Luo,He summed up the pathogenesis of NAION luo-blood deficiency and stasis resistance,then founded Gui-Yuan Ti Luo Yang Xue prescription.For the the previous clinical curative effect,we are going to discuss the effect and mechanism of Gui-Yuan Ti Luo Yang Xue prescription on NAION.ObjectiveTo observe pathological change for different doses of Gui-Yuan Ti Luo Yang Xue prescription on NAION rats’optic nerve.To explore the improvement and mechanism of apoptosis that Gui-Yuan Ti Luo Yang Xue prescription on NAION rats’s retinal ganglion cell(RGC).Also the improvement and mechanism of vascular contraction and oxidative stress that Gui-Yuan Ti Luo Yang Xue prescription on NAION rats.MethodsDivided 96 healthy rats into 6 groups randomly as normal group,model group,mecobalamine group,low dose traditional Chinese medicine(TCM)group,medium dose TCM group and high dose TCM group.NAION rat model was prepared by photodynamic therapy.Three doses of Gui-Yuan Ti Luo Yang Xue prescription were given to the corresponding rats by gavage for 28 days.The mecobalamine was given to rats in mecobalamine group by gavage for 28 days.Distilled water was given to rats in modem group by gavage for 28 days.The normal group did not intervene.The changes of Gui-Yuan Ti Luo Yang Xue prescription on NAION rats’RGCs were observed with fluorescence microscope photographs,HE staining,transmission electron microscopy,western blotting,enzyme linked immunosorbent assays,radioimmunoassay and colorimetric method.Results1.Observe the HE staining picture of optic nerve,normal group rats’ optic fibers arranged orderly with glial cells.After 28days,model group rats can be found optic atrophy,which retinal nerve fiber layer weakness,and hyperplasia in optic nerve,rythropoiesis.Mecobalamine group rats can be found optic atrophy,the obvious loosened optic nerve fiber layer structure.Low dose TCM group rats can be found optic atrophy,reticular irregular retinal nerve fiber layer,Medium dose TCM group rats can be found optic atrophy,loosened optic nerve fiber layer structure.High dose TCM group rats can be found the orderly optic nerve fiber layer structure,and minor loosened optic nerve fiber layer structure.Observe the HE staining picture of retinal structures,norrmal group rats’ retina structural integrity and have a rich number of RGCs.After 28days,model group rats can be found that RGCs were lost a lot,mecobalamine group and Low dose TCM group rats can be found that RGCs were lost.Medium dose TCM group rats were much better,High dose TCM group rats were the best.Observe the nerve transmission electron microscopy results,normal group rats’optic nerve fiber bundles vary in size.The optic nerve fiber bundle has a large number of neatly arranged axons.After 28days,model group rats’ optic axons disappeared in large Numbers,the glial scar is obvious with axon swelling,myelin collapsed.mecobalamine group rats’optic axons disappeared a lot.The glial scar is obvious with axon swelling.Low dose TCM group rats’ optic axons disappeared a lot.Which was similar to the previous group.Medium dose TCM group rats’optic axons were slightly disordered,part of axon is missing.High dose TCM group rats’optic axons are arranged in an orderly manner with myelin abnormal occasionally.Observe the fluorescence microscopy,normal group rats’ RGCs form clearly and radial distribution.Model group rats’RGCs number were obviously down,Other groups’better.Survival rate of RGCs:There were statistically significant differences between the groups.The other groups were statistically significant with the normal group.It was statistically significant compared with the model group for 3 doses of TCM group and mecobalamine group.And it was statistically significant compared with the mecobalamine group for medium dose group and high dose group.The high dose TCM group and the medium dose TCM group were statistically significant.2.Caspase-3 in different groups had statistical significance.It was statistically significant compared with the normal group for 3 dose TCM group and mecobalamine group.It was statistically significant compared with the model group for high dose TCM group and mecobalamine group.And it was statistically significant compared with the mecobalamine group for medium dose group and low dose group.The low dose TCM group and the high dose TCM group were statistically significant.The medium dose TCM group and the high dose TCM group were statistically significant.Caspase-9 in different groups had statistical significance.The other groups were statistically significant with the normal group.The high dose TCM group and the modern group were statistically significant.The high dose TCM group and the mecobalamine group were statistically significant.The high dose TCM group and the low group were statistically significant.The high dose TCM group and the medium group were statistically significant.Bcl-2 in different groups had statistical significance,The other groups were statistically significant with the normal group.The high dose TCM group and the model group were statistically significant.The high dose TCM group and the mecobalamine group were statistically significant.The high dose TCM group and the low group were statistically significant.The high dose TCM group and the medium group were statistically significant.Bax in different groups had statistical significance,It was statistically significant compared with the normal group for low dose TCM group and model group.It was statistically significant compared with the model group for 3 dose TCM group and mecobalamine group.And it was statistically significant compared with the mecobalamine group for low dose TCM group and high dose TCM group.It was statistically significant compared with the low dose TCM group for medium dose TCM group and high dose TCM group.The high dose TCM group and the medium group were statistically significant.Fas in different groups had statistical significance,It was statistically significant compared with the normal group for high dose TCM group and mecobalamine group.It was statistically significant compared with the model group for high dose TCM group and mecobalamine group.It was statistically significant compared with the mecobalamine group for low dose TCM group medium dose TCM group and high dose TCM group.The high dose TCM group and the low group were statistically significant.The high group for high dose TCM group and normal group.NO in different groups had no statistical significance.ET-1 in different groups had statistical significance,The other groups were statistically significant with the normal group.It was statistically significant compared with the model group for low dose TCM group medium dose TCM group and high dose TCM group.It was statistically significant compared with the low dose TCM group for medium dose TCM group and high dose TCM group.The high dose TCM group and the medium group were statistically significant.Conclusions1.Gui-Yuan Ti Luo Yang Xue prescription played a positive role in repairmen of NAION pathological organization form,It can delay the death of RGCs,The high-dose group worked better.2.The high dose Gui-Yuan Ti Luo Yang Xue prescription may be able to inhibit the exogenous pathway and endogenous pathway protein apoptosis,activate apoptotic inhibition protein expression,and inhibit the apoptosis of RGCs.3.In the treatment of NAION,The high dose Gui-Yuan Ti Luo Yang Xue prescription has the function of maintaining vascular tension,keeping organ irrigation,helping vascular stability and repairing damage.The high dose Gui-Yuan Ti Luo Yang Xue prescription can inhibit the accumulation of ROS and promote the SOD,reduce the generation and accumulation of lipid peroxides,delay the adverse effects of oxidative stress,maintain balance between oxidation and anti-oxidation effect of methods to protection of RGCs.
