Objective:Through the comparative study for 3 months,6 months,12 months male and female mice endogenous metabolic marker changes and the callback function on metabolic markers of four tonic prescriptions in different months,the paper revealed the biological basis of four tonic prescriptions and syndrome associated with the different aging stage biological basis.Methods:Experiments were made to select 3 months as the control group of youth,females and males are both 10.1206 months old mice and 120 12 months old mice with half males and half females were divided randomly into 6 groups:Sijunzi Decoction group,Siwu Decoction group,Liuweidihuang Decoction group,Jinkuishenqi Decoction group,Vitamin E group and blank control group with 10 mice in each group.Five drug groups were treated with four tonic prescriptions and Vitamin E,and the rest of the two groups were filled with the same volume of physiological saline.In the experiment of the 3 1th day,every mice was taken eyes to gather blood and separated plasma,killed the mouse and retained the spleen,After liquid nitrogen frozen samples,immediately turned them in the freezer for deposit.Metabonomics analysis were made on the methanol extract of the plasma and spleen,and three factors in correlation analysis on different months of age,gender,medication group were discussed based on metabonomics results.Results:(1)Compared with 3 M group,there were 28 spleen metabolomics markers in 6 months female mice.They were associated with four synthesis,metabolic pathways.Four tonic prescriptions and vitamin E had a callback function to the above 4 way s.(2)Compared with 3M group,there were 26 spleen metabolomics markers in 6 months male mice.They were associated with 21 synthesis,metabolic pathways.Four tonic prescriptions and vitamin E had a callback function to the above 6 way s.(3)Compared with 3 M group,there were 8 plasma metabolomics markers in 6 months female mice.They were associated with 8 synthesis,metabolic pathways.Four tonic prescriptions and vitamin E had a callback function to the above 8 way s.(4)Compared with 3M group,there were 22 plasma metabolomics markers in 6 months male mice.They were associated with 17 synthesis,metabolic pathways.Four tonic prescriptions and vitamin E had a callback function to the above 13 ways.(5)Compared with 3M group,there were 34 spleen metabolomics markers in 12 months female mice.They were associated with 27 synthesis,metabolic pathways.Four tonic prescriptions and vitamin E had a callback function to the above 23 ways.(6)Compa red with 3 M group,there were 22 spleen metabolomics markers in 12 months male mice.They were associated with 19 synthesis,metabolic pathways.Four tonic prescriptions and vitamin E had a callback function to the above 4 way s.(7)Compared with 3M group,there were 6 plasma metabolomics markers in 12 months female mice.They were associated with 7 synthesis,metabolic pathways.Four tonic prescriptions and vitamin E had a callback function to the above 3 ways.(8)Compa red with 3M group,there were 9 plasma metabolomics markers in 12 months male mice.They were associated with 6 synthesis,metabolic pathways.Four tonic prescriptions and vitamin E had a callback function to the above 4 ways.Conclusion:Compared with 3M group,spleen and plasma metabolomics markers of 6 and 12 months male and female mice,whose synthesis and metabolic pathways were all in a certain change associated with sugar metabolism,lipid metabolism,amino acid metabolism,nucleotide metabolism and other important material metabolism,and four tonic prescriptions and vitamin E had certain callback function to changes in the above ways.
