measure to prevent the occurrence of TURP syndrome in advance.This is still a clinical problem.Endoscopic monitor has been used as a noninvasive tool for irrigation fluid absorption and blood loss assessment of transcervical resection of endometrium(TCRE)and transcervical resection of myoma(TCRM).During useing Endoscopic monitor examine the clinical predictors of decreased risk of fluid absorption and blood loss of hysteroscopic procedures.And by recording relevant biochemical,promote the recovery of postoperative homeosasis.Endoscopic monitor has clinical practical value.Methods:Endoscopic monitor based on the method of the strain gauge transducers,the photoelectric sensor and volumetric fluid balance.The system used to detect the inflows of the irrigating solutions,outflows of the irrigating solutions,fluid absorption and bleeding.Thirty TCRE and thirty TCRM consecutive patients undergoing were investigated over a 14-month period.The volume of intra-operative fluid absorption and blood loss was measured using Endoscopic monitor.We also need to record the Change of heart rate,pulse oxygen saturation,Hb,electrolytes pre-and post-operation.Data were analysised by Statistical.Results:In TCRE,the irrigation time is 18-47min(34.7±8.7min),irrigation fluid volume is3164-8935ml(7134.7±1834.4ml),the volumes aborbed is129-397ml(300.5±80.6ml),the volume of blood loss is 20.9-57.9ml(34.4±9.9ml),Pressure,endometrial thickness and uterine depth have positive correlation with fluid absorption and blood loss.The data of electrolytes Na~+had significant difference in TCRE(P<0.05),Na~+ have negative correlation with fluid absorption and blood loss when the operation time is more than forty minutes.In TCRM,the irrigation time is31-70min(41.1±7.5min),irrigation fluid volume is 6116-14789ml(7855.2±1743.1ml),the volumes aborbed is 254-876ml(415.2±113.4ml),the volume of blood loss is 65.8-188.9ml(101.0±24.0ml).Pressure and uterine fibroids size have positive correlation with fluid absorption and blood loss.The data of electrolytes Na~+,Cl~-and CO2-CP had significant difference in TCRM(P<0.05),Na~+ and Cl~- have negative correlation with fluid absorption and blood loss when the operation time is more than forty minutes.Conclusion:Endoscopic monitor is a simple and efficient method to measure fluid absorption and blood loss.It could be used to detect potentially dangerous and to improve the safety of operation.Shorten the operation time can reduce fluid absorption and blood loss.When operation time more than 40 minutes will cause the change of patients with electrolyte,so prevent the happening of the TURP syndrome in advance.Objective:In genetic subtype and tumor microenvironment epithelial ovarian cancer and basal-like breast carcinoma have many similarities,so these two tumors may exist same drug treatment.Lycorine is a isoquinoline alkaloid,and has extensive antitumor effect.The aim of this study was to research the antitumor effect of lycorine on SKOV3 and MDA-MB-468 cells in vitro and to further illuminate the mechanism of autophagy and apoptosis,migration and invasion and discuss the possible mechanism of joint action.Methods:We used MTT to assess the cell viability of SKOV3 and MDA-MB-468 cells,with different concentrations of lycorine for 24 h,48h and 72 h.Application the flow cytometry technology,we researched the variation trends of cell cycle disposed by 25ug/ml lycorine for 72 h exposure.Using Hoechst33258 and Monodansylcadaverine(MDC)staining observed the cell multiple of apoptosis rate and autophagy body.Crystal violet staining was used to detect migration and invasion.We further tested the apoptosis and autophagy protein(Bax,Bcl-2,PI3 K,AKT,PAKT,P53,P62,LC3 B,ATG5 and caspase-3)and migration and invasion proteins(MMP2,MMP9)in SKOV3 and MDA-MB-468 cells.Results:In different time,different concentrations of lycorine suppress SKOV3 and MDA-MB-468 cells were not same,inhibition rate was positively correlated with time and concentration.We analyzed that lycorine blocked G0/G1 phase of SKOV3 and G2/M phase of MDA-MB-468 cells through flow cytometry.Besides,the multiple of apoptotic rates in 25μg/ml lycorine treated SKOV3 cells for 24 h,48h and 72 h were(1.32±0.108),(1.61±0.098)and(1.77±0.087)(P<0.05),the multiple of apoptotic rates in MDA-MB-468 cells were(1.43±0.13),(1.74±0.14)and(2.15±0.14)(P<0.05).Two kinds of cells have the typical autophagy bubble formation.Crystal violet staining indicated that lycorine decreased migration and invasion.Western immunoblotting confirmed that Lycorine significantly decreased the expression of Bcl-2,but increased the expression of Bax and promoted the caspases 3.Furthermore,lycorine upregulated the expression levels of LC3 II and ATG5 along with downregulation of P62 in SKOV3 and MDA-MB-468 cells,two markers for autophagy.Lycorine induced the activation of PI3K/Akt /Pakt protein kinase in skov3 and MDA-MB-468 cells,accompanied by upregulation of p53.In addition,lycorine decreased MMP2 and MMP9.Conclusion:The proliferation of SKOV3 and MDA-MB-468 cells were inhibited under lycorine,the present study indicated that lycorine could induce autophagic and apoptosis by inhibiting commonly PI3
