OBJECTIVE:① To investigate the relationship between the number of respiratory department outpatients and the number of PM2.5,PM10 and AQI in the Affiliated Hospital of Shandong University of Traditional Chinese Medicine.② To establish animal models of inhaled granules in laboratory,observe the different preventive and therapeutic effects of three prescriptions of Puji Xiaodu Drink,Xuefu Zhuyu Decoction and Yupingfeng Powder.METHODS:The mice were placed in a special exposure box,let the smoke generated by coal combustion enter into the exposure box for modeling,the PM2.5 concentration maintained at 700-900ug/m3,to establish inhalable particles exposure models.The decoctions of three prescriptions of Puji Xiaodu Drink,Xuefuzhuyu Decoction and Yupingfeng Powder were respectively administered to the exposed models orally,by the way of prophylaxis,prevention+treatment and treatment.Observate lung pathological changes under microscope,and changes of tracheal mucosa cilia by electron microscopy scanning.The proportion of dust cells in the alveolar lavage fluid was counted.The number of CD68+ macrophages was observed by immunohistochemical method,the ratio of CD4+and CD8+ T cells was detected by hemorheology.The concentration of IL-1β,TNF-α,NF-κB and TRPV1 in lung tissue homogenate and were concentrations of serum IgE and IgG in serum detected by ELISA.And the expression of NF-κB mRNA and CD68mRNA in lung tissue was detected by PCR.RESULTS:The results of clinical study showed that the female was more sensitive to the respiratory reaction of haze,outpatients to 50-69-year-old majority.The outpatient visits reached the peak in 10,11 and 12 months.The influence of pollution factors on the incidence of outpatients in respiratory department was the most obvious at 2 weeks postponement.PM10 had less influence on the total number of patients and the age groups.The concentration of PM2.5 had the greatest influence on the number of patients in respiratory department,and had the greatest influence on 30-49y crowd.② In oxidative stress,the Qingrejiedu prescription is useless of anti-oxidation mechanism of prevent,single-blood circulation prescription and Yiqi prescription have a certain prevention use,the three prescription in curing the body Due to inhalation of particles caused by oxidative stress have a certain role,and sustained drug effects of the best intervention.③ In the aspect of cellular immunity,the there prescriptions may be have different effect the time node of macrophage function,and the effect of prescription of promoting blood circulation and removing blood stasis is the best.On T cell function,Xuefuzhuyu decoction and Yupingfeng powder prevention using could improve the immune function of T cells to a certain extent,and maintain in the toxic environment.On Single treatment use,the effect of Xuefuzhuyutang is better than the other two prescriptions.④On humoral immunity,Yiqi prescription application can improve the body’s humoral immune function,but no better than Qingrejiedu prescription and the type of blood circulation when the mice has been exposed in polluted environment already.⑤ On inflammatory cytokines,the effect on IL-1βof Qingrejiedu prescription and blood circulation prescription was better.The effect of Yiqi GuBiao prescription on TNF-a was better,Yiqi GuBiao Recipe and activating blood circulation Stasis prescription on the NF-κB intervention stronger.In the three prescriptions,the effect of activating blood circulation and removing blood stasis drugs on inflammatory cytokines was the strongest compared with other two kinds of prescriptions.⑥ Direct response to physical and chemical stimuli,three prescriptions can reduce the physical properties of respirable particles on the lungs of direct stimulation,thereby reducing airway hyperresponsiveness.Horizontal comparison,regardless of preventive medication or treatment,are the role of Yupingfengsan more prominent,compared with the other two prescriptions.CONCLUSION:The effect of PM2.5 on the respiratory system of the population is not only acute pathogenic effect,but also cause chronic long-term damage.Chinese medicine have a certain degree on the human body against inhalation particles,the effectiveness of different prescriptions are different,so applicate Chinese medicine to prevent and treat respirable particles caused by respiratory damage can not rely on a single effect Prescription,syndrome differentiation at different stages,and long-term use can allowing Chinese medicine to play a greater role. |
